An introduction to on-section CLEM - Duration: 11:59.Correlative light and electron microscopy methods (or CLEM methods) allow us to take advantage
of the information we receive from the light microscope and combine it with the electron microscope
to precisely localise a protein of interest within a tissue.
Here we're going to take you step by step through a on-section CLEM technique
that has been developed in the labs of John Briggs and Marko Kaksonen.
This method combines the good ultra structural preservation given by high pressure freezing
and free substitution, and allows us to preserve the fluorescence in a methyl acrylate resin.
The advantage is is that we can do the light microscopy on a thin section
that has been placed on an electron microscopy grid.
This gives us a very good resolution in Z and allows us to go directly
from the light microscope to the electron microscope to do the imaging.
This method has proven to work very well on several biological models,
ranging from cultured cells to tissues.
The limitations being the size of the specimen for good effective freezing,
but also the level of the initial fluorescent signal.
So depending on the sample you're working with some steps may have to be adapted.
In this example we are using a monolayer of HeLa cells expressing a GFP-tagged protein
that is found in the Golgi apparatus.
In preparation for high pressure freezing we grow the cells on a 3 millimeter
sapphire disc that has been coated with a 10 nanometer layer of carbon.
High pressure freezing is performed with small metal carriers where one side has
a 300 micron well and the other side is flat.
The carriers are first put into a petri dish on a piece of filter paper coated with hexadecene.
First we replace one carrier with the flat side facing up into the high-pressure freezing holder.
The sapphire coverslip bearing the cells is then transferred cells face-up on to the carrier.
It is important at this stage that no air bubbles are introduced.
A gold spacer is placed on top of the sapphire disc, followed by another metal
carrier with the flat side facing the cells.
The sandwich is finally tightly secured within the high-pressure freezing holder and placed into the
high-pressure freezing machine for freezing.
This apparatus first applies 2,100 bars of pressure onto the sample
and then quickly freezes by a jet of liquid nitrogen.
The cells are thus vitrified within 20 milliseconds.
After freezing, the holder is promptly transferred to a container filled with
liquid nitrogen, where the carriers can be dislodged from the holder and stored in a small Eppendorf tube.
For free substitution, we're using the Leica AFS-2 machine plus the FSP unit.
The machine is cooled down and the solutions are put
into the machine to let them cool down before transferring the samples.
Using a binocular we transfer the carrier sandwich in an Eppendorf filled with
liquid nitrogen, and place it into the machine.
We then separate the sapphire disc away from the carrier and the spacer ring and place it into the
free substitution cocktail with the cells facing up.
At this point you should make sure the sapphire disc is laying flat in the cocktail and not at an angle.
Once all sapphire discs are transferred, the FSP unit is attached and plugged
into the machine with the syringe lined up and ready to go.
The machine usually runs between 5 to 7 days, depending on the free substitution protocol you choose.
During the free substitution process the samples are incubated with 0.1% uranyl acetate in acetone.
This percentage of UA is a compromise between obtaining a nicely stained sample
without quenching the fluorescence.
The initial incubation in the free substitution cocktail is an important one, and certain samples
can benefit from lengthening or shortening this time period depending on how thick they are.
For cell monolayers, we normally incubate them in the free substitution cocktail for 9 hours.
After this incubation period the cells are rinsed with acetone followed by a step by step infiltration
with Lowicryl HM20 resin.
Once the blocks are polymerised, the holder is taken out and the blocks are removed
and can be stored for months at room temperature in the dark, until they are ready to be used.
For the sectioning we are using an ultramicrotome and sectioning with an ultra 35 degree diamond knife.
You can choose which thickness to section depending on your electron microscopy imaging strategy.
For 2D imaging we cut 70 nanometer thick sections and for 3D electron tomography 300 nanometers.
Sections are retrieved on a 200 mesh carbon coated copper grid.
We like these grids particularly because they are stable and have an asymmetric mark in the centre
that can be used as a reference in the microscope,
and the likelihood of the structures of interest lying on the grid bar is minimal.
Once collected, the grids are transferred to a box where they can be protected from light.
The fluorescence signal is very sensitive and grids must be imaged within the same day to avoid bleaching.
To perform the correlation we use beads that can be seen both in the light microscope and the electron microscope.
These are commercially available beads that are electron dense and fluorescent,
and can be used for this purpose.
Here we use TetraSpecks that can be seen in the blue, green, red and far-red channels.
These TetraSpecks can be bought in different sizes, but for this application it's important to choose the
smallest ones in order to increase the precision of the correlation.
Here we incubate the grid with the sections facing down on a 10 microlitre drop
of TetraSpeck solution in PBS.
The concentration of the bead solution is determined empirically prior to the experiment.
As we will see later, we will aim for a dense enough distribution to detect dozens of beads
on a single field of view at the electron microscope.
After 5 minutes incubation the fiducial beads are rinsed with distilled water.
The grids are blotted with filter paper in between each washing step, and placed back in the storage box.
For imaging in the light microscope we sandwich the grid between two glass cover slips,
so the grid is flat and as close to the objective lens as possible.
First place a 30 microlitre drop of water on the first cover slip and place
the grid on top with the cells facing down.
In order to avoid the formation of air bubbles we pipette some water on top of the grid to wet the surface.
We then put a thin layer of vacuum grease around the edge of the second
cover slip and carefully place it on top of the grid.
Close the sandwich, pressing very gently around the edges to spread the water.
To mount the sandwich on the microscope we insert it into a custom-made imaging chamber.
For the light microscopy we typically use a wide-field fluorescence microscope.
For imaging we put the imaging chamber onto the microscope stand
Using the 20x objective in the bright field mode we retrieve the centre of the grid
which contains the asymmetric mark for orientation.
Switching to the channel of interest, we acquire an image with a very low excitation intensity and exposure,
in order to localise the cells of interest without bleaching out the signal.
Next, we save the position of the cell and change the objective to one with a higher magnification.
In this case we chose a 100x oil objective with a 1.4 NA.
We go back to the cell of interest and we focus on the beads using a channel
that does not excite the fluorescent protein of interest, and take an image.
We then switch to the channel of the fluorescent protein of interest and take the image.
To remove the grid from the coverslip sandwich, unscrew the chamber
and open the two cover slips using a sharp scalpel.
The grid, sections and carbon film tend to stick to the surface of the coverslip
so it's important to treat it gently and remove it without damaging it.
Place a drop of water onto the grid and carefully pipette up and down
until the grid releases from the surface.
Take the grid and blot off the excess liquid with a filter paper.
The grid is then imaged in the electron microscope and we can use the asymmetric
mark in the centre to navigate to the cell of interest.
Once we reach the cell, we acquire images at different magnifications to later be used for the correlation.
A lower magnification can be used to locate enough fiducials for the correlation,
and a higher magnification for higher resolution images of the areas positive for fluorescence.
For the correlation we are using the IC software package and the eC-CLEM plugin.
In the EM image the fiducials will appear as dark electron dense spots, 100 nanometers in diameter.
While in the light microscope image they appear as diffraction-limited spots positive for the four channels.
In this example we can see green and red.
We select corresponding beads by clicking on their centre.
It's important to use the same channel as the protein of interest to select the centre of the beads
to avoid shifts due to the chromatic aberration.
After localising 3 beads in both imaging modalities the software is able to calculate the transformation.
However the more beads you click the more precise the correlation will be.
The software applies a similarity transformation that scales rotates and translates the fluorescent image
to make it fit to the EM image.
This results in a final image overlay where we can zoom in and appreciate which structures
underlie the fluorescent signal.
In this case we see a clear representation of the Golgi apparatus

little bit the next thing was a I was being a rapper I told everybody I knew I
wanted to be a rapper and you know I was just a colossal filler I was not good
I could not ride the beat at all but I had a lot of fun you know I'm asked a
lot of I'm asked equipment saved up so I can't I don't have money for equipment
yeah was gonna save up you know took me I think a year or a couple years
to get everything I didn't get everything I want all the equipment at
once but yeah it was not a good rapper and I never made a dollar off and all I
did was spend money spend money I wasn't even smart enough back then to
make it as a hobby I'm a tax return but I wasn't really making anything more
taxes who cares anyways yeah I failed at that too but like I get an idea in my
head and I want to do it was it a total failure I don't know because look I
never really stopped doing anything I think I'm gonna come around I'm gonna
drop a ha ha drop a hot Jim but it taught me that you know I kind of have a
knack for getting the equipment together I guess being a sound engineer or a
producer I really I think I have an ear for like who's good and stuff so but in
my in my future I definitely wanna have like a recording studio maybe be an
engineer and and sell recording time or just do a recording time with my friends
again and so I jumped on an idea I went for it I mean it was a total failure I'm
sure I'm sure there's a ton of people well no I mean I'm sure there's some
people going head dumb ass you know it's not I could be a rapper he he showed him
he sucked you know but it's like yeah ok they can say all that
I mean I I hope there's some people that how I feel about it was at least I tried
at least I didn't just watch from the bleachers you know like I thought hey I
love rapping I loved the art for I want to do that and so I tried and it wasn't
for me I mean I but I learned some other stuff that is for me and what I'm saying
is to failures professional professional athlete and rapper both failures
both ideas I had that I wanted to be I don't care it doesn't bother me and
shouldn't bother you to fail because then you've got someone else you know I
would think if I knew somebody said they want to be a rapper and they actually
tried for a little bit and then moved on to something else I wouldn't laugh at
them for trying I think oh that's cool I would maybe get inspired to try my own
thing well after after the rapping I never really said I would you know stop
that but anyways I wanted to be a certified public accountant with my own
business and that's what I wanted and that was a dream I had that was an idea
I had I went for it I didn't let the other maybe our failures maybe help me
because I've went for other stuff before didn't get it
life goes on but at least I tried the difference here is between the other two
times I really believed in myself I started believing myself the other two I
don't know why I would even announce I really wouldn't announce to everyone
in the world that I was a rapper but I don't know why weird announce it to
anybody if I never believed in it myself I don't know if I announced that I would
be a professional sports player that's maybe the problem is I didn't announce
tutor because I didn't believe in myself but I believe that could be a CPA and I
announced it to everybody and that was a big idea a big dream and I went for it I
got it because I'm not afraid to fail and you shouldn't be either another
another and so I got my CPA and then another step was you know opening my own
business now I've been opened that was another idea crazy idea but I did it now
I've been open for two and a half years and I'm turning away clients
this YouTube thing of course this YouTube thing even after all those
failures but then I had a couple wins but I said hey I don't care I want to do
the e time i watch the each of them I wanted to youtube so I tried the YouTube
and now so I've given you two things I've been failures at and I guess two
things three things I've been successful at I am happy that I tried all five
things you know it's a learning experience I'm I'm upset that I didn't
try more stuff or really give it my all but you know it feels so good so
liberating to have an idea and just go for it just go for your dreams you know
just try it just try it one time just try it one time you know there's five
different things that you know people told me oh you can't do that thing no
one was like oh that's a great idea except being a CPA of course that was it
that was the one thing out of the five that was a good idea although when I
said I wanted to quit to do YouTube so I could YouTube and lift more and then
make a and and make a little bit of money yeah everyone thought that was
crazy but that's what I that's what I that's the idea that I had in my head
right and I want to act on my ideas because you asked you definitely you
asked an older person you know they're more they're not sad about what they
tried you make it or you don't and and you can change your ideas and and not
like it but when you look back it's it's what you haven't done
you know knowing and and and there are going to be some people say you say you
start something up but I think it's the fear of failing most other people don't
really want to see you do better so they'll tell you oh yeah it's hard don't
do that you know a lot of people are like oh no he'll leave us or something
contentment life anyways yes sir there gonna be some people that talk mess but
you don't need to worry about those people it's I don't know why I see I
have this analogy in my head that only only one set of exercise will work good
for you yes there's gonna be certain like did there's gonna be certain people
say no um only only the exercise that I do is good and so they'd make fun of
somebody for doing a different exercise you know I wouldn't make fun of anybody
for exercising just the same just the same way as I wouldn't make fun of fun
of anybody for going with their idea you know trying to get a pad and trying to
get their idea selling a couple ideas so um and whatever your idea is just make
it happen just try it so much it's so much fun
just think of mmm think of all the people that watch football on Sundays
like but versus the amount I've actually played it's almost like life like um
most of people that I know that played football in high school or whatever
played football don't really watch it all day long some do but me because
that's they're bitter or something or they have other things to do but you you
you could look at the amount of people that play football versus like
professionals what a thousand maybe versus a
millions and millions and billions of people who watch I probably wouldn't
extrapolate it that much to other life but you know how many people actually
participate in in life and with the internet anybody you know you can do
anything these days you can learn anything do anything there's no excuses
you know there's no excuses in this day and age you know you you can go to the
library and get on the Wi-Fi and make things happen you can do that for free
you can go to a library anywhere in the US so I don't I don't know what anyways
excuses but you you know get off get off the get off the bleachers and go play
that's what I'm saying it's so much more fun you know cuz yeah like I gave you a
a couple big fails in my life and then I've given you some successes and I
don't know I just this fails I just who cares
but the successes are awesome so it's like shooting shooting a three-pointer a
basketball I love shooting three-pointers in basketball and you
know when I'm on a runway and I'm hitting feels good and when I'm missing
I gotta have amnesia but the thing is you're not you're not gonna it doesn't
feel the best when you miss but hey it's it feels so good when you win so step
out of the bleachers and get on the field what's your idea what's your
what's your thing that you think you can't do what's your thing that your
family's gonna say it I don't do you then I don't do that all your friends
yeah don't do that you know I'm just you know you got to live for yourself but
anyways run with your idea cuz your idea is nothing without some action this is

In the aftermath of the devastating Columbine high school tragedy, America began searching
for someone to blame — and narrowed in on one person in particular.

At the time, Manson was already seen by Middle America as the antichrist.
When word got out that the shooters were goths who had listened to his music, the nation
went nuts.
In an interview with The Guardian, Manson later revealed,
"But, honestly, the Columbine era destroyed my entire career at the time."
That being said, he claims to understand why he was a target.
"Because I think it's easy to throw my face on a TV because I'm in the end the poster
boy for fear."
His concerts were protested and he received hundreds of violent threats.
The most aggravating part?
As Manson later wrote in an article for Rolling Stone,
"Responsible journalists have reported with less publicity that [the shooters] were not
Marilyn Manson fans – that they even disliked my music."
Michelle Shocked
In 2013, singer Michelle Shocked fired off some hateful anti-LGBT comments while playing
a club in San Francisco.
According to The Guardian, Shocked told the crowd,
"Once prop 8 gets instated, and once preachers are held at gunpoint and forced to marry the
homosexuals, I'm pretty sure that would be the signal for Jesus to come on back...If
someone would be so gracious as to please tweet out 'Michelle Shocked just said from
stage, God Hates F------.''"
Shocked had been a successful artist, nominated for an MTV Video Music Award alongside Madonna
and Sinead O'Connor back in 1990.
The controversy left Shocked at the center of a media storm and on the wrong side of
her own fans.
In a long interview piece with the singer, Dallas Morning News claimed in 2017 that the
outburst sent Shocked's career into a near-terminal nosedive, losing her bookings and wiping out
a planned tour.
Lee Ryan
In October 2001, a wounded America was still trying to understand the events of September
11th, as the rest of the world watched sympathetically — with one exception.
Lee Ryan of the British boyband Blue was not impressed with all the grief and solidarity.
That month, in an interview with the Sun, Ryan asked,
"Who gives a f--- about New York when elephants are being killed?"
Ryan went on to say,
"They are ignoring animals that are more important.
Animals need saving and that's more important.
This New York thing is being blown out of all proportion."
Fittingly for those who believe in karma, Ryan and the rest of Blue split a couple years
later, eventually going bankrupt.
The Dixie Chicks
In 2003, The Dixie Chicks were one of the biggest acts in country music.
Until, that is, they took to a stage in London, where lead singer Natalie Maines declared
to the crowd her dislike of then-President George W. Bush.
"Just so you know, we're on the good side with y'all.
We do not want this war, this violence, and we're ashamed that the president of the United
States is from Texas."
As The Guardian detailed, that was the exact moment the Chicks' career hit the red, white,
and blue fan.
Country music dropped the all-girl outfit like radioactive waste.
Their singles plummeted down the Billboard charts, radio stations banned their music,
and they received so many violent threats that they were forced to install metal detectors
at their concerts.
The Chicks' sales never recovered.
B.o.B
In January 2016, hip-hop artist B.o.B decided to take "retro" to the next level by taking
geography back to the Stone Age.
In a tweetstorm, he fired off pictures and messages designed to prove that we've all
been lied to about the shape of the Earth — and that it's actually flat.
The series of tweets has come to define what's left of B.o.B's career.
His subsequent Twitter spats with guys like astrophysicist Neil deGrasse Tyson have sent
him down a rabbit hole that has since consumed his entire life.
In September 2017, B.o.B even launched a GoFundMe campaign to raise $1 million to launch satellites
into space to "find the curve," an impressively obtuse way of saying "to prove the Earth is
flat."
It doesn't appear this battle will end well for the rapper-turned-explorer.
Sly Stone
The collapse of Sly Stone's career was less a single moment than a whole series of desperate
moments that finally culminated in one uber-moment that effectively ruined his reputation forever.
After stuffing hefty amounts of stuff up his nose, the singer started missing concerts
and collapsing on stage.
As People detailed in a 1996 profile, his pet dog mauled his son in 1976, his wife left
him, and his band broke up.
Still, Stone could have probably recovered from all this.
But the one thing his record label took a much dimmer view on was Stone getting busted
for possession.
After that, he didn't work again for over a decade.

Kevin Mannis purchased it at an estate sale since then various strange
happenings have befallen its owners to list just a few it smells of cat urine
or jasmine flowers lights burn out randomly and strange
health problems regardless of the stories told by its different owners
some believe these individuals were already looking for bad stuff to happen
and as such they believe they've been cursed when they actually start

like any good paranormal mystery this doll has been causing a lot of
conversation it first came to light when Debbie from King's Lynn in Norfolk UK
claimed that the doll had scratched her husband yes that's right the doll is

the story doesn't stop with Debbie instead of doing the good thing and

but of course she did the next owner Lee steer from rather omim South Yorkshire
one that tense international bidding war to own the doll at a costly one thousand
one hundred sixty two dollars after raising the money with some of his
paranormal investigator friends after bringing the doll home Lee's father soon
began experiencing the same issues as Debbie's husband it appears the doll
doesn't like married men overall both Debbie and Lee believe the scratches are
the same size as the dolls hand this is supposedly one of the most expensive
cursed and haunted objects sold on eBay hopefully Lee and his family decide to
destroy the doll and not sell it again number 7 the hands resist him painting
this painting has been the subject of many urban legends and it is believed to
carry a haunting curse created by artist bill Stoneham it is believed to have
been associated with the death of several people like any good haunted or
cursed object once it went up for sell on eBay in 2000 many enthusiastic
paranormal object collectors started bidding on it selling for $1,025
hopefully the unlucky buyer hasn't faced any ill fortune or hasn't been haunted
by some unknown entity number six the Bassano vase this beautiful vase dates
from the 15th century Italy and is believed to be the subject of a terrible
curse not much is known about its exact history but the legend states it was
created for an Italian bride who lived in a village close to Napoli
unfortunately on the night of her wedding the bride was found lying on the
floor dying and clutching the vase close to her before death could finally take
her the bride promised she would return and seek her vengeance after her death
the vase was passed from one member of the family to the next and strangely all
are reported to have died under very mysterious circumstances finally someone
had the sense to lock it away in some believed it was buried on sacred ground
by a priest what ever happened to it though it resurfaced in 1988 when it was
then attributed to the deaths of three more people a pharmacist a surgeon and
an archaeologist so where is it today well nobody knows for certain it is
believed that someone finally decided to do something about it though so
hopefully it won't be unearthed again any time soon number 5 the Delhi purple
sapphire as far as famous gems go the Delhi purple sapphire is not
particularly remarkable it was set in an unattractive silver ring with alchemical
and astronomical signs since it has never actually been sold it has never
been given a monetary value but should it appear on the black market it would
likely sell for more than a thousand ironically although the piece is
referred to as a sapphire it is in fact an amethyst gem so where did it come
from well like the quarry nor diamond which
we'll cover later in this list it is believed to have originated in India
where it is believed to have been looted from the temple of Indra during the
indian mutiny of 1857 it is also believed that through the theft of the
item from the temple of the Hindu god of war and whether that a curse was cast so
what is the curse well it certainly didn't bring any good fortune to Colonel
W Farris who brought the gem back to England soon after his return his family
faced many financial and health problems and then things took a turn for the
worse when a family friend committed suicide while in possession of the gem
the next poor victim to fall foul to the curse was author Edward Aaron Allen who
had suffered misfortunes and bad luck that made him believe it was terribly
cursed ironically Edward was not a man known to believe in mythology or
superstitions so it's even more interesting that he changed his mind
about the gym his fear over the curse was so strong that upon the birth of his
daughter he locked a gem away in a vault and upon instruction following his death
he gifted it to the Natural History Museum with strict instructions not to
open it for three years after his death and that his daughter was never allowed
to touch it in a letter that accompanied the gem he finishes with a warning of
the gems curse and that it should be cast into the sea unfortunately his
advice was not acted upon and the jewel is still in the museum's possession
today number four a Syrian artifact value just over 1 million dollars this
Assyrian artifact is a steely dated about 2,800 years old forming a part of
a larger piece which depicts King a da dinner re the 3rd of Assyria this piece
has a curse engraved directly on it that's convenient but it's not just the
curse that's been causing problems when antiques dealer Halima Corbin at
tempted to sell the artifact at auction police officer soon raided the warehouse
and seized the object after being informed that it was likely stolen and
brought to Geneva illegally although korban does not seem to have been
affected by the curse itself it definitely hasn't brought him any good
luck number three the cursed watch purchased by sheikh bin mohammed al
thani a Qatari royal in 1999 for 11 million dollars
this watch is believed to have caused many issues for its previous owner and
later for the Sheikh himself so what's its history well it was commissioned by
an American businessman Henry Graves and created by Patek Phillipe who it was
subsequently named after this is where the strange happenings begin in the
months after taking possession of the watch Henry graves best friend dies and
months after that his son died in a car crash but this isn't the end of the
mysterious cursed watch although it remained dormant for some years it was
later purchased by Sheikh bin Muhammad Al Thani who died a mysterious death in
his London home in the hours before he managed to sell it to some unknown buyer
thankfully the cell managed to settle some of the debt the Royal had incurred
but unlucky for the buyer perhaps we might soon hear the next development in
this story number two the Kohinoor diamond the
Kohinoor diamond is a massive gem that was originally 186 carats until Prince
Albert ordered it to be cut down to 105 point 6 carats today it is embedded in
the UK Imperial state crown worn by female consorts like the late Queen
Mother it is valued at about 125 million dollars in today's money so what's the
story behind it and why is it believed to carry a deadly curse for any male
owner well it was originally mined in India around 1100 CE e and remained in
the country until the East India Company annexed the Punjab after the anglo-sikh
Wars prior to this the diamond passed through the hands of many
and several of its Mel owners were all violently dethroned as a result it is
now believed to carry a curse against mill owners and that only women or God
can wear it with impunity thankfully no ill fortune has fallen upon the British
monarchy well unless you want to include the collapse of the British Empire
number one the Hope Diamond I'm sure you knew this was on the list but number one
is the Hope Diamond probably one of the most famous jewels in the world and
there is a record of its ownership dating back nearly 400 years
weighing forty five point five four carats it's famous for its rare blue
color caused by its chemical makeup today it is housed in the National
Museum of Natural History in the United States so what's the controversy well in
the late 19th and early 20th century it was believed the original stone was
stolen from a sculptor of the Hindu goddess Sita although this is also a
legend believed to have been started by Western authors during the Victorian era
nevertheless there are numerous accounts of people being affected by ill fortune
and bad luck some of those supposedly affected are Jim merchant John Baptiste
Tavernier the story states that he came down with
a raging fever after stealing the diamond and died then again other
accounts say he lived till he was 84 King Louie the fourteenth who died of
gangrene and all but one of his legitimate children died in childhood
Dutch jeweler Wilhelm falls he recut the diamond and ended up being murdered by
his own son Greek merchant Simon monka writes he drove his car over a cliff
killing himself and his family James Todd the mailman who delivered the
diamond to its new owners the Smithsonian apparently had his lake
crush shortly after and his house burned down now these are only a few examples
of the stories associated to this cursed diamond and in all more than 13 deaths
are linked to the possession of the Hope Diamond well they all might be
coincidences they definitely - the mystery of this object today many
non-believers in the curse think the stories were just fabricated or
exaggerated to increase the value of the jewel

Right now, if you were to go to a reseller like an OTA,
and buy a ticket to a tourist attraction,
whether it's a hop-on-hop-off bus ride,
an observation deck, or a museum,
they don't give you a real ticket.
They give you a paper voucher.
You have to stand in line and exchange that
for the real ticket.
But then, the real problems happen
for the tourist attraction after that.
They have to count them, verify them,
and send them back to get paid.
Redeam eliminates all of that process
and creates an electronic link
between the reseller and the supplier
such that, as soon as you book,
it's already in the reseller's system.
And when you show up,
you just scan your ticket and go inside.
- [Interviewer] So this has been a challenge
for a lot of the travel industry for a long time.
And a lot of people have had a go at trying to fix it,

Which is just crazy, in, you know, today's day and age.
And so we create the system that allows the attractions
to operate a lot more in a modern fashion.

just the networking that you can do here,
the kind of people that attend Phocuswright.
- Yeah, the networkings great.
I mean, it's one of the few places where you can find
the CEOs of the largest OTAs
just wandering around the halls.
And you can stop and be like, oh, hey,
you know, I want to talk to you about something.
And you can't do that a lot of places.

Police in Iraq say a suicide bombing at a popular market on the outskirts of the capital,
Baghdad, has killed at least 11 people.
Officials say at least 26 others were wounded in the attack, which took place in the Nahrawan
shopping area southeast of Baghdad.
A security official said two suicide bombers opened fire on civilians in the market late
Monday, then blew themselves up.
The Islamic State [group] terrorist group claimed responsibility for the attack in a
message on its Amaq channel.
Myanmar's military chief said he told Pope Francis that there is "no religious discrimination"
in his country, where violence against Rohingya Muslims has been termed "ethnic cleansing"
by the United States.
[General] Senior General Min Aung Hlaing said in a Facebook post "Myanmar has no religious
discrimination at all.
Likewise our military too... (it) performs for the peace and stability of the country."
Pope Francis met with Myanmar's military chief Monday as he began his trip to the southeast
Asian country to discuss violence in Rakhine state which has caused over 620,000 Rohingya
Muslims to flee into neighboring Bangladesh.
Thousands of Myanmar's nearly 700,000 Catholics traveled to greet the Pope as he landed in
Yangon Monday, and more than 150,000 have registered to attend a Mass he will say on
Wednesday.

Turkish officials say they are watching to see whether the United States will honor a
promise, which they claim was made by President Trump to Turkey's president last week, to

Artist Vlog #2: Difference Between Reality Dream And Unrealistic Daydreaming | Oil Painting Process - Duration: 13:21.hello everyone, it's Lavennz here and welcome to today's video.
And today I'm going to talk about this painting that I have completed and vanished the week
before I go to Pasadena and Los Angeles so if you remember a film partially of my last video,
the Sketchbook Tour, in the hotel room I stay in Pasadena
so right now I'm back to Maine since Tuesday and
I'm kind of miss those wonderful moments I have in Southern California
I love the food there, the scenery, the weather, the unique houses I saw around Pasadena
which I never saw this kind of building somewhere else but maybe in Mexico or
Southern America which I haven't visited yet and I also did some
watercolor landscape sketches in my travelers notebook and some travel diary
so this is the mountain view I saw from the windows of my hotel room
based on the map this mountains is supposed to be call as
Angeles National Forest
and I was really inspired by the view to
use the cold earth tone the paint the rock and the mountains and the skies
and I'm definitely going to use these color scheme in my next video
so the second sketch is a sunset sky view.
I'm not very good in watercolor landscape painting so
it looks rather like ocean or something abstract not cloud and sky
so that's just two sketches I did there
and I also spent a day in Universal Studio Hollywood which is so much fun
and I love the studio tour around Hollywood and also the 3D rides yeah
that's amazing and I love the feeling they create in their Harry Potter world.
I don't have a good picture because I didn't bring my
cameras to the park I just want to enjoy have fun there
but well, this is what it looks like based on the plastic bag from the souvenir I bought.
I only bought two souvenir there for myself so the one this one is what I'm wearing, the Gryffindor scarf.
Well, it looked like a typical scarf if you're not the Harry Potter fan
but I am so, I know this is Gryffindor scarf and I also yep here's the bag
I also bought a magic one from Ollivanders
The wand I choose is the great wizard, Professor Dumbledore
it comes in the box. It's a collectible artifact
And this is what Dumbledore's wand look like
I'm not going to play with it or maybe I will when there's no one around
yeah but this is something I'm interested to collect and I keep it in my studio with me
I was thinking that maybe I would do a painting with a witch or wizard and a wand in the hand
so this would be the perfect props for the painting reference photo.
so let's get back to today's topic which is this painting
I call her 'Her Little Paradise'
and this is the first ever painting I do on darker
skin tone and I already show you the portrait study of the painting in September video
so I take a new approach in painting this piece which I'm going
to show you in the painting time-lapse later in these video
basically I do a lot of wet on wet technique in the painting background and I have two
important secret ingredient, magic ingredient, that make the background
shine and sparkly and reactive with light you can see I hope you can
see it I see the purple here is more vivid when there's a light shining on it
and when you turn, err, it's not so easy to see but in in real life you will notice the difference and
special ingredient is I already show you in my previous videos which is the interference powder pigment
the one I Jacquard Pearl Ex Pigment Powder and I'm using the green and violet
The pigment powder itself is very pale that you can
hardly see the colors on it but when you apply this on the darker background
the colors will come out vividly very saturated especially when they
reflecting the light
so I really love the sparkly starry background this the
sparkly starry background of the painting is very fantasy which is
actually the purpose of this background in the painting
is to represent the unrealistic the the fantasies of this lady which
mean is something she dream of
and to bring the painting balance in between fantasy and reality
I also add some laundry earring, the hanging laundry earrings from her ears
well I know it's weird but I don't know I just have this idea like
something just hanging on our head to remind us, hanging around ears to remind us
what's the reality needs and how to do to survive and to me
laundry is one of the best thing to represent reality because this is
something that we don't, nobody love to do laundry and I know
even my mother who is very clean and tidy but I don't think she enjoyed laundry as well
so and me myself don't, but we have to do it right otherwise we'll ran out of clothes to wear
unless you're very rich, you just buy one and throw it away after you wore which is a waste
add a house on top of the lady, is
represent something that she dream and could be a reality if she approaching it
so basically this painting is about dream
so to me, dream is something's come up from our fantasy and we try our best effort to bring it to reality which
mean making the dream comes true thus
without actions, it's not a dream. It's daydreaming
and so as we must always that
I feel that we must always remember
what we truly want, our honest desire the rooted deeply in our heart and we
have to like take one step at a time in approaching this dream to bring is the reality
and when it happened we are living in
our own little paradise that built with our own hands so
well that's the basic ideas, the basic concepts of this painting I have
but if I'm going to go into the detail, each of my ideas of this painting
this videos were going like last, I don't know, maybe hours if I can talk too much
Yeah I'm not usually can talk so fluently and honestly I actually have to write up
some what I want to talk before I actually firming it because I always, my mind
always go blank so I guess I have to like train on public speaking right
