BREAKING: Obama's Nasty UN Plot Exposed | Top Stories Today - Duration: 3:39.

When one thinks of the United Nations, one can imagine the leaders and ambassadors of

the world gathering to discuss what is happening on a global scale.

This particular UN meeting was slightly different.

Despite former President Obama retiring from the White House and moving on with his life

as a private citizen, he decided to make an appearance at the UN, according to CNN.

During his time there, he managed to completely undermine President Trump and speak at an

event as if he still were in the Oval Office.

All other presidents have stayed away and let the new representative of the country

speak without detraction.

After all, his two terms are up, right?

So what makes Obama an exception?

Liberal media outlet CNN even noted that Obama spoke "In a speech and question-and-answer

session that closely resembled the types of appearances he favored as president."

The language suggests that he is, at least to some degree, thinks he's still President.

The former President had a chance to speak at Bill and Melinda Gates Foundation's "Goalkeepers"

event.

He managed never directly to call out President Trump or Republicans by name, but the words

that he used said it all.

Obama described the efforts to repeal and replace ObamaCare as "aggravating," despite

the fact that it is a failing system that people no longer want.

He proudly touted that people were alive today thanks to the Affordable Care Act.

He failed to mention the people who saw a tripling — if not more — of their coverage

premiums.

He never once apologized to the minimum wage workers, that he swore he wanted to protect,

who lost their coverage and ended up paying a deductible that consisted of all of their

tax money, or close to it.

Sen. Bill Cassidy (R-LA) and Sen. Lindsey Graham (R-SC) have been working to push a

repeal of the Affordable Care Act.

President Trump has already agreed on numerous occasions that he would sign it, but it has

yet to come to fruition.

"Now, the legislation that we passed was full of things that still need to be fixed.

It wasn't perfect, but it was better," said Obama.

Obama went on to clarify, "So when I see people trying to undo that hard-won progress

for the 50th or 60th time, with bills that would raise costs or reduce coverage or roll

back protections … It is aggravating.

And all of this being done without any demonstrable economic or actuarial or plain common-sense

rationale, it frustrates."

It may be hard for him to understand, but the American people have spoken.

ObamaCare has done more harm than good; there is no doubt about that.

It is time for a change.

Since President Trump's election, the Left–and specifically Barack Obama's operatives–have

attempted to undermine his presidency.

Is the Left trying to derail the Trump agenda?

Obama then stated that he was frustrated with the fact that he had to gather people every

couple of months to keep "leaders" from inflicting "real human suffering."

Even without saying names, it is crystal clear what he is getting at here.

The previous attempt to repeal and replace ObamaCare occurred when Republican Senators

John McCain (R-AZ), Shelley Moore Capito (R-WV) and Lisa Murkowski (R-AK) joined the Democrats

and voted against the bill that would end ObamaCare once and for all.

In doing so, they turned their back on the wishes of their constituents — Americans

who voted for President Trump and his promise for repeal.

I hate my natural hair - Duration: 7:40.

Hi guys! In today's video we're going to talk about

the way we view our hair

Disclaimer : my channel is about natural hair so I'm always going to push this agenda

But if you have relaxed hair or think about relaxing it, no problem! Who am I to judge ?

So I'm going to talk to you about my own experience, how I went from regarding my hair as an iron bullet

around my ankle to now, seeing it as one of my striking feature

I've not always liked my hair, actually, I hated it when I was younger

I really didn't like it

It had nothing to do with self hatred

Or not being able to accept myself

It was practical, I didn't like my hair because I had extremely thick and frizzy, coily hair

And it was difficult to tame it (let alone take care of it)

For me, the styling proces was just a torture

And when my mother would take her purple comb (it was a fine tooth comb)...

I would die a little inside

All I wanted as a little girl was to have straight hair

Hair you could detangle without having your comb hitting a knot every 2 seconds

Without having to pull my hair to remove all the knots

...even if it meant tear off my head at the same time

I just wanted the styling proces to be shorter and less painful

But I don't blame my mother: she always did what she could with what she had

And she did a great job given the compliments we received about our hairstyles, my sister and I

So when did my mind changed about my hair ?

Well, it changed when I started to ask myself the right questions

When I stopped wondering how to straighten my hair (it really didn't help me)

And started to think "how can I care for my hair as it is?"

(Why don't I have long hair ?!)

(this was actually the main question)

It really gave me the keys of the gates to the natural hair world, they opened, I jumped in...

And I never came back

From then, I discovered lots of things and experimented with my hair

And did things I didn't think were possible with it

And I acquired more self-confidence

That's why I think here lies the key if you have a hard time accepting your hair

Ask yourself the right questions

Ok, you don't like your hair but what don't you like about it ? Be more specific

Is it its length, is it too dry, is it damaged, is it its texture ?

For most of the questions you have, you can find answers if you're ready to look out for them

You also need to be able to see yourself in a positive light and acknowledge the things

you like about your hair

Like its volume or its curls

And these are the things you want to keep, the things you want people to see

As I said it, it's easy to find solutions if you look out for them, there's a lot of information

Because everything is just one click away with Internet

But you need to stay critical because you find so much information and advice

You'll see lots of people telling you to do something and then the opposite

So you need to be able to sort what is going to be useful for you and what is not

And adjust it to your needs and personnality

Because most of the people you see, you watch on YouTube

(I include myself in those people)

Except if they are professionals like hairdressers

Can only talk about their own experience and it's, of course, different than yours

So, again, you need to be wise about what you see and hear on the Internet

Don't compare yourself to anybody you see, especially not to people

Who have different type of hair, different texture

Why ? Because it means their hair has different needs

And sometimes it's really frustrating when you have extremely frizzy, coily hair, you'd like it to be curly

Because we think it looks better and it's easier to take care of it

But having these kind of thoughts regularly

Get your in a negative state of mind

You're going to be very frustrated

And rough with your hair

Try to find some kind of model, a person who has hair that looks like yours

Because it's going to create some kind of connection

And you'll see that there are people with your type of hair who "succeed"

But also that that person, if she's honest, and shows you the good and the bad,

also has problems sometimes, just like you

Sometimes she feels frustrated about her hair, she might want to give up, her hair comes out the wrong way

So you'll see that this person also struggles

Just like you

And it will make your life easier (not trying to be mean)

Because you then see that we're all on the same boat

And it's always better to suffer together than suffering alone

I also want to say something to mothers here who might have daughters with natural hair

Teach them to like their hair it's extremely important

Tell them how much of a good thing this hair can be!

Even if it's different from what they typically see [on TV]

Show them that they can do many things with if, there are plenty of possibilities!

It's unlimited!

And that it can be really fun to play with it

So that they don't think that taking care of their hair is a chore

But regard it as an activity that makes it possible for them to take care of themselves

And they'll also have more self-confidence

I give you all this tips but don't forget that the acceptance is not going to come at all once;

If you really want to work on that and learn to embrace your hair you need to be patient

It doesn't happen in one night

You don't wake up thinking :

I loooooooooooooove my natural hair!

No!

It doesn't happen like that

Again, you need to be patient, let yourself (and your hair) some time to mature that

And be gentle with it

Because when we're angry, when we're upset we tend to be rough with it

And it won't help you to like it

But if even after all this questionning, you still can't stand your hair

Then do what you have to do with it!

The goal is to feel good about yourself

And this mane of yours

hey guys what is going on just maniacal here and i am back with another dead by

daylight video it's been a little while last one I did was of the Huntress and

I'm going back to the trapper cuz he's probably one of my favorite I don't know

I really liked the Huntress the only problem is that I don't really have any

practice on her so I'm just going back to what I'm good at

or better at than the rest so just keep you guys up to-date here's my loadout

I got brutal strength this one I'm not gonna try to pronounce it

well basically 12% faster when I'm carrying everybody I believe and then I

get to see people when they're down within 12 meters so yup thats about it

Canon Camera Remote Control RC-6 Review + Demonstration | JEN TALKS FOREVER - Duration: 4:12.

and just like that we're rolling hi I'm Jen I want to talk about this gadget

today because it has changed my life I don't know why I didn't start using it

earlier I bought it like two years ago and for some unknown reason I just

started using it like yesterday which was a huge mistake I guess I like to

make my life harder so this is a Canon remote for cameras Canon cameras it's

the rc6 I got it on Amazon I think it's about 25

to 27 dollars and for some reason I bought it and just kept it in a box

until just this past weekend so I thought I would get it out and start

using it and oh my gosh what a game-changer so often if you're a

vlogger or a youtuber you know you film alone a lot you're a one-man band you

know you're doing it all you're trying to set up your shot and get everything

ready you have to try to take pictures of yourself for your thumbnail so I

don't know why I didn't start using this remote before now I guess I forgot I had

it or something but this thing can make it so that you can hit the button and

start recording stop recording and you can also take photos of yourself so I'm

gonna do a little bit of a demonstration of this product obviously I recommend it

because I think this thing is freaking amazing and it's very small it comes

with a little case how it works is you have to set up your camera and put it

there's a button on top that says dry so what you do alright let me turn on this

camera and I have two cameras I've got the Canon 70d and the Canon a DD

obviously I'm shooting with the a DD so I'm using the 70d so you just find the

button on top that says drive and then you want to just make sure see if a few

different options you can do either at the 10-second remote or the two-second

remote I choose to do the two-second remote I don't need that long to take a

picture so all you do is select that there's a few different things that you

can do so on the back of the remote there's a dot so you can either click

this down for the dot or click this up for the two if you want to use the

remote for video recording it has to be in the two position if it's in the two

position but you're taking still photos it will give you that self timer mode if

it's on the dot it will actually immediately take the picture and then

all you have to do is make sure that your camera is facing you so perfect for

selfie mode right so just make sure it's facing you and then all you have to do

now there's a chance I may accidentally turn off this recording hopefully not

let's see oh I do have this set on - all right so now I'm recording over here and

all I do do is press the button if I want to stop all I do is hit the against

I'm trying not to stop this recording because I don't want to do that and

that's all there is to it it is so easy and this way like if I have a shot set

up I have a sewing YouTube channel as well I don't have to keep getting up and

sitting down to start and stop recording so I think that's pretty super awesome

so yes I am loving this Canon remote it's the RC six I've linked it below if

you want to pick one up there are quite a few knockoffs that are cheaper I can't

really speak from personal experience so if you want to go from one of those you

are taking a risk just know that off the bat but this one is awesome

and I think for the price I think it's well worth it if you liked this video

Republican Lawmaker Fired After "All Lives Splatter" Facebook Post - Duration: 3:31.

South Dakota majority whip, Lynne Disanto, has lost her job.

I should say, her day job with Keller Williams Realty over the fact that on Facebook Disanto,

again Republican whip for the House of Representatives for the state of South Dakota, posted a meme

of somebody running over protestors, and said, "Nobody cares about your protest, get your

ass out of the street, #alllivessplatter."

Disanto then added her own caption that said, "This is a movement I think we can all get

behind, #alllivessplatter."

Now the real estate company, Keller Williams, had enough sense of duty, enough morals, and

enough courage to tell Disanto that this was disgusting.

We're not going to employ you anymore, so once your tenure is up in Congress, don't

bother coming back, because we don't want anything to do with you.

Now as for Republicans in the South Dakota Legislature, they're perfectly fine with Disanto

posting a meme advocating the murder of protestors here in the United States, and keep in mind,

when this meme was originally posted by Disanto, it was less than one month after somebody

in Charlottesville did just that.

They ran their car over protestors and killed someone.

Republicans all over this country are trying to make that legal by passing legislation

saying that if you're in a street, and you're protesting, we can legally run you down and

there's nothing you can do about it.

And Disanto thinks that, that's funny.

Thinks that, that's a good thing.

Thinks it's a movement we can all get behind.

And then later on when she was frigging pressed about this, she said, "Oh, I meant it more

as a stay out of the street public awareness type thing.

This wasn't meant to be some kind of horrible ..."

Here's the exact quote here.

Let me say this.

"I perceived it as encouraging people to stay out of the street."

That's all it was folks, it was just a public service announcement gone awry.

Don't go out into the street, because you could get hit by a car, you know we all tell

our kids that.

But that's not what you thought, you lying piece of crap.

You knew what it meant, otherwise you wouldn't have put the #alllivessplatter, and you wouldn't

have shared a meme that says, nobody cares about your protest.

You are a piece of human garbage.

And the fact that the people in South Dakota elected you to represent them, doesn't speak

too highly on them either.

Hopefully now that they see your true colors, they will have enough sense to vote your ass

out of office next time you come up for re-election, because it's people like you that give this

country a bad name.

It's people like you that make other countries laugh at us, and it's people like you that

are trying to squash dissent of much more intelligent people than yourself, who are

trying to enact real change here in the

United States.

adventure and you want Disney to share in that adventure not with Mickey Mouse

not with Donald Duck not with goofy or Pluto this is a

How Phasma Got Away With Murder - Duration: 13:27.

What's up Star Wars Fans?

My name is Prince, and I am an Urban Acolyte.

Delilah Dawson's Phasma novel gave us the backstory on Captain Phasma, and it was really

interesting how her origin story is presented.

Vi Moradi, a smuggler and a spy working for Lei's Resistance is captured by the First

Order.

She's being interrogated by a high ranking First Order officer named Cardinal, and their

interrogation delves into Phasma's backstory.

What we learn is that Phasma is hiding some secrets that could halt her quick ascent into

the upper ranks of the First Order's Chain of Command and eventually bring about her

undoing.

So I want to talk about that big secret in the Phasma novel and tell you all how Phasma

got away with Murder.

Before I get into all that, I just want to say that if you're new here, I talk about

Star Wars, martial arts, and how to train like a Jedi in the real world.

Allright--before I go any further with this video, I should tell you that I am going to

discuss SPOILERS from the Phasma novel.

Some of the stuff I'm about to say may carry over into Star Wars Episode 8, the Last Jedi,

so if you're trying to avoid spoilers, this may not be the video you want to be watching,

okay?

I'm just saying, I don't want to see anyone crying about how I spoiled the book or the

movie for you if you're watching beyond this point.

So as I was saying earlier, Cardinal has beef with Phasma, big beef.

Go back and watch yesterday's video where I asked if Phasma may be Force sensitive.

At the end, I mentioned that Cardinal may have been one of the children who left Jakku

on Palpatine's Naboo yacht with Brendol and Armitage Hux and Grand Admiral Rae Sloane,

at the end of Empire's End after the Empire made it's last stand at the Battle of Jakku--

this dude Cardinal was actually born on Jakku, so there's something to store in your memory

banks for later when I discuss Cardinal and his possible importance to the Resistance.

Maybe something that could come up in Episode 8.

I told you guys some of this stuff could pop up in a movie later.

So I'm saying all of this because Cardinal had been part of the First Order from the

very beginning.

He became Brendol Hux's right hand man-- he appears to be the first stormtrooper to have

officially been given a name that wasn't some serial number like TK-237.

He also wore red armor and a cape, and you know--if you get a cape, in Star Wars, you're

basically hot stuff.

So we know that Cardinal's position didn't last.

Phasma was brought in by Brendol Hux.

She quickly rose through the ranks.

She never had to do all the propaganda programming, and she soon became Cardinal's equal.

Where he was once responsible for all of the stormtrooper training, He now only worked

with the children and Phasma was given the teenagers and young adults who were about

to graduate from their training phase.

So this dude Cardinal is looking to take Phasma out--and that's where Vi Moradi comes into

play because she's got the scoop on Phasma.

She knows every secret that Phasma doesn't want anyone to know--or so we think.

So recall how Phasma impressed Brendol Hux on Parnassos-- if you don't know what I'm

talking about, just go back and check out yesterday's video.

Well, as it turns out, Phasma didn't only impress Brendol Hux, she also impressed the

younger Hux, Armitage, who we know as General Hux.

Now recall that Armitage was Brendol's bastard from what I think was an extramarital affair

Brendol was engaged in while running the Imperial officer cadet program on Arkanis.

Somehow he managed to retain his position even after Ezra, Sabine and Zeb tore up this

academy when they went to rescue Zare and his Force sensitive sister, Dhara, Leonis.

To be honest, I think that event could have pushed Brendol's head even further up his

backside, creating the pompous jackass that we saw in the Aftermath series and in the

Phasma novel.

I mean, Hux was probably a genius when it came to training young people to serve the

Empire and later the First Order, but this guy...he's like that fat drill sergeant who

can get their recruits to admire them until they find out what a HUGE DOUCHEBAG they really

are when they aren't on duty.

Armitage and Brendol didn't have a good relationship at all-- we saw that when Rae Sloane basically

had to tell Brendol that if he ever touched his son again that she'd break his legs or

space him or something like that.

Although that probably ended the physical abuse, the neglect continued.

Like I said, Brendol was the kind of guy to command respect from his troops, but treat

his family like dirt.

This guy Cardinal was basically, the son that Armitage was not.

He seemed to enjoy spending more time with Cardinal than with Armitage, and Armitage

was only viewed as like, this instrument of his legacy.

Anything Armitage accomplished in life would be seen as like an extension of Brendol's

greatness.

Now you're probably thinking, "Prince, this is supposed to be about Phasma, why you talking

about Cardinal, Brendol, and Armitage Hux?"

Well, like I said, Phasma managed to impress not only Brendol Hux, but she also impressed

Armitage Hux.

Phasma had a lot of ambition, and even though she swore her allegiance to the First Order,

Phasma appeared to be willing to do anything, kill anyone, to push her agenda...and we don't

really know what Phasma's agenda is.

Is it self preservation?

Is she after power?

We don't know what Phasma's real goal is, and that makes her a pretty scary individual.

What we do know is this...I said Phasma is willing to kill anyone, and the title of this

video is "How Phasma got away with murder."

Phasma killed someone in the First Order-- a high ranking officer who, at one time, was

important to Palpatine's contingency plans to build a new Empire.

On Parnassos, where Phasma was born, in the desert wastelands, there are these blood beetles

who live beneath the surface.

These blood beetles will infect anyone they bite some kind of toxin that will cause a

person to become extremely inflamed in a short period of time, like in a few days to the

point that they basically burst into liquid and what's left of their guts.

Picture what happened to Senator Kelly in the first X-Men movie when Magneto alters

him so that he has the mutant gene.

He exploded into a puddle of water in the floor when his cells became unstable.

So I'll cut to the chase-- Phasma infected Brendol Hux with one of those blood beetles..."after

everything he'd done to help her reach her position within the First Order."

She killed him in a way that would leave no evidence.

He burst into a bunch of liquid and guts inside of a bacta tank.

The First Order had never come across this kind of creature, and they had no way to identify

any toxins that were present in what was left of Hux.

All anyone knew was that he came down with flu-like symptoms and died of unknown causes.

Now the story gets even more interesting because when Vi Moradi told all of this to Cardinal,

he confronted General Hux, and get this--it was actually Hux who had Phasma kill his father.

Hux worked with Phasma to have his father removed--and thus securing Hux's position

within the First Order and moving him up the Chain of Command and closer to Supreme Leader

Snoke.

By reporting everything he knew to General Hux, Cardinal actually brought about his own

demise.

He was seen as someone attempting to undermine whatever this power base Hux had created with

Phasma's help.

Eventually Phasma confronted Cardinal, and she nearly killed him in a fight where she

completely managed to outclass him at every step.

The crazy thing is this-- Phasma not only killed Brendol Hux, the man who helped her

reach her position, but we also learned that Phasma orchestrated the murder of her parents

when she was in her adolescence, and just before leaving her homeworld with Brendol

Hux and her neice, she killed her own brother after defeating at least 30 people armed with

only a blaster and a spear.

Phasma is a cold-blooded killer, and she will turn on anyone who gets in her way.

Anyone who sees Phasma's face or knows her history before the First Order basically has

a death mark on them, in Phasma's mind.

I asked if Phasma could be Force sensitive, and someone replied that if she were, Snoke

and Kylo Ren would sense it and train her.

That's a cute assumption, but why would you train someone already gifted to become even

more powerful?

Kylo Ren and Snoke may not be Sith, but I don't think they would want to share their

power the same way a Sith would not--they would not want to empower someone to become

a rival.

If Phasma did have the Force, and she learned to harness it, I could easily see her attempting

to take down Kylo Ren, and Snoke, so that she could rule the First Order herself.

Does this sound like me making a huge leap?

Maybe, if you're not thinking about what we've already seen Phasma do-- think about it now--why

did Phasma lower the shields to Starkiller Base with so little resistance?

If the base and the planet are destroyed, but Phasma can find a way off the base--Both

Kylo Ren and General Hux could be killed.

It would leave 2-sets empty in that triumvirate of power and make her the top dog in the First

Order under Supreme Leader Snoke.

We know, or at least anyone who read the Phasma book, that Phasma has some kind of internal

compass, where she always knows where she is and where she's going.

We're seeing in the Phasma comic book series exactly how she escaped the planet-- so I

think Phasma had a plan when Finn, Chewie and Han showed up demanding she take down

the shields.

I think her plan was to allow the base to be destroyed and see if she could use that

opportunity as a power grab.

Like I said, Phasma got away with murder before.

She murdered her entire family.

She murdered General Hux's father, Brendol Hux, one of the most important people to the

start and rise of the First Order.

I don't think Phasma will have any trouble dropping enough bodies to help her get whatever

it is that she wants.

She's already got a long track record of getting away with Murder, so why not keep going?

But that's all I've got to say for this video, so I'd like to hear from all of you.

the mask?

I said in yesterday's video that she's showing signs of the same traits that Rey has.

Could you see Phasma eventually rising to the point that she's in some super high position

with the First Order?

I mean, honestly, I would not be surprised if Phasma turns out to be the actual Darth

Vader of this story moving forward--and what I mean is that she could be the person who

has some kind of big reveal as the story progresses-- so could you see something like that happening,

and what do you think that big reveal would be?

Don't say that you think she'll be Rey's mother because I'm getting ready to debunk that over

the weekend, so check back for that video, if you are holding on to any hope of her being

Rey's mother or having anything to do with how Rey ended up on Jakku.

When they come to drill team, we basically let

them know that they have about 6 months to reach

their first major milestone.

If they are not able to accomplish that, then

platoon.

That type of dedication is something you can't teach.

What can I say?

As soon as I arrived 1 year ago he was someone

who initially stood out to me that took initiative

He's looked at what was on my plate and has always

asked me what I can do to help and how can I be an

added value to the drill team.

He's actually one of the top qualitied members

which mean that he's not just one of the leaders of

drill team but he's one of the leads of this command.

In all honestly I was the underdog.

It was kind of depressing.

At first I didn't really see it because back in

high school I wasn't, I don't want to say bullied

but I definitely wasn't the most popular kid.

There was some point that I did think I can't do

this anymore but it all goes back to my

grandfather.

He told me way back when that I could do whatever

you need to do, just do it.

So I would just listen to that and I would just call

him as much as possible and tell him , hey I'm

doing better, and then he died, so I was like you

know what I'm still going to do this and I'm going

to do this for him.

So every time we do our prayers before we go out

there I make sure he knows that I'm up in front now.

RECONSTRUCTING THE BRAIN IN A DISH

EMERGENCE OF NEUROINFLAMMATION FROM RETROTRANSPOSONS

In this work we took advantage of a rare disorder

like Aicardi-Goutieres to really explore the fundamental mechanism

that happens in your brain.

Clinically you can see the manifestation

pre-birth by ultrasound.

You can detect that their brains are not developing well.

They have microcephalic brains.

Overall, it's a very severe neurological disorder.

DESIGNING THE STUDY AND CHOOSING TARGETS FOR INVESTIGATION

There are a couple of things that we knew about Aicardi-Goutieres.

One is the fact that it behaves like an autoimmune response

in the absence of any obvious infection.

Second is the nature of the genetic mutations

in genes such as TREX1.

And finally, the complete absence of an animal model,

they don't show the neurological abnormalities

that we see in those patients.

STUDY TARGETS: TREX1 AND LINE1 ELEMENTS

TREX1 stands for three prime repair exonuclease 1.

Exonuclease in general, they degrade RNA or DNA,

so the body can defend against a virus.

So TREX1 basically its function is to degrade DNA,

single-stranded DNA.

If you look at TREX1 inside the cells,

we see an upregulation of repetitive regions of the genome,

including retrotransposons such as line retrotransposons.

Line 1 elements are repetitive sequences

that are able to move from one region of the genome into another

by a mechanism called retrotransposition,

and the way that they do that is by using reverse transcriptase.

As far as we know, Line 1 elements are active during early development

and also in the brain.

CHOOSING A METHODOLOGY: STEM CELLS

With the absence of a good animal model,

the only way to study this condition in a human background

is to take advantage of human pluripotent stem cells.

So we use IPSCs, induced pluripotent stem cells,

and embryonic stem cells.

And you create specific mutations and use these cells

to differentiate specific cell lines.

Using these cell lines

we are able to create a different model,

an organoid, that recapitulates

the process that we have in our brain development.

MODEL FUNCTION AND OBSERVED EFFECTS

So when we engineered the cells

we confirmed that the mutation is there

but there was no TREX1 protein inside those cells.

In a healthy cell you always have your DNA,

in the nucleus, where it's supposed to be.

And so what we saw is that outside of the nucleus

to an extreme amount, single-stranded DNA existing.

And a lot of the DNA was actually the Line 1 retrotransposon element.

They behaved like normal cells

with the TREX1 protein being expressed.

The big surprise came when we pushed or instructed

these neuro progenitor cells to become neurons,

and we see a dramatic effect.

By a large proportion, the AGS patient cells were just dying.

We knew then that it was a problem of cell-death.

The TREX1 mutant cells virtually cannot make neurons.

That strong dramatic phenotype prompted us to make astrocytes.

And when we compare the amount of Line retrotransposons

we realized that there were way more

in the cytoplasm compared to controls.

What we see is that these cells look like immunoreactive.

They feel like they were attacked by a virus or something, we think.

They basically sense retrotransposons as the enemy

and they trigger this interferon response.

The astrocyte signals, they were toxic.

So AGS patients have intrinsic toxicity

within the neuron itself,

and extrinsic toxicity from the astrocytes.

So they're getting hit twice.

EFFECTS ON ORGANOIDS

In the 3D system the effects are even more dramatic.

The main thing we saw on the AGS side

is that they were dying as fast as they were growing.

And as a consequence we have cerebral organoids

that looks like microcephalic.

They're smaller in size and the amount of cell death

that we see in those neurons

is elevated compared to the control cells.

THERAPEUTIC OPPORTUNITIES:

REVERSE TRANSCRIPTION AND ENDOGENOUS CELL TOXICITY

Once we have these dramatic effects

we thought about a potential therapeutic intervention.

So there are actually reverse transcriptase drugs.

So we tried these same drugs.

When you added these retrovirals,

there was much less single-stranded DNA in the cytocell,

and then we saw that they were much healthier.

So they didn't have the toxicity.

They didn't have the interferon response in astrocytes.

We completely rescued the cell death.

Virtually they become indistinguishable from the controls.

In the brain organoids it works as nice as in the 2D system.

The retroviral drugs could actually make these brain organoids

thrive as much as in the control cells.

THERAPEUTIC OPPORTUNITIES: CONTROLLING INTERFERON RESPONSE

Essentially astrocytes are secreting interferons

that are killing neurons.

If you could block this pathway

you could prevent the extrinsic cell death.

To do that we actually use a drug

that can block the interferon receptor,

and by doing that, we could see the same effects.

We rescue the amount of neuronal death

that we see in the mutant cells

to a level that's comparable to the control cells.

CONCLUSIONS AND IMPLICATIONS OF THE STUDY

The conclusions of our work is that we could actually

nail down the exact mechanism

both at the molecular and the cellular level

of what's going on during the neurodevelopment

of Aicardi-Goutieres syndrome.

So we are definitely excited with the idea that one can design

a clinical trial to treat Aicardi-Goutieres.

But it's also possible that this mechanism

is more fundamental than we think,

and it might actually be working or contributing to other diseases

such as autism, aging, or neurodegenerative disorders

where Line elements are highly expressed in their brains.

So it might be more common that we think.

Anyone can be a scientist if they wanted badly enough if they work really hard

and if they are really excited about what they're doing. My name is Angie

O'Neill, I'm a senior instructor here at UBC in the Department of Zoology.

Growing up everyone always told me, "oh you're a smart kid you should be a doctor" and I

was interested in the human body I was interested in learning about how that

works, so I thought, yeah that would be kind of neat. But then when I went to

university, I all of a sudden learned about all these amazing careers that you

could have as a scientist, you know being a doctor wasn't the only science career

you could have. When I was a grad student I got to TA a course for the first time

and I just fell in love with teaching. It was the most exciting thing ever. And I

found that every, everyday I'd be thinking about it even when I didn't

need to, I was thinking about, oh how am I going to teach my students is how am I

gonna ask them and get them excited about learning about this. And I found

that I was just thinking about it when I didn't need to and I always think if

you're thinking about something when you don't need it's a sign that it's

probably something you want to do with your life. The first time I ever got to

teach a course as faculty, it was a last-minute cancellation and this was a

Vertebrate Structure and Function course. I've never taught the course

before and I didn't really know the material, the person who regularly taught

the course was away so I didn't know where anything was stored, and I was so

excited! Because this is my chance and I really wanted to do a good job, but since

I didn't know all of the material I basically had to work seven days a week

eighteen hours a day. I was so tired I would fall asleep sitting up taking my socks

off. I thought this is my chance and I want to do an amazing job of this.

I think it's not worth doing something if you're not gonna do it really well so I

just I went for it and I put my whole heart into it and and gave it my best.

And I think if you're willing to put that amount of effort into it and make

it such a big part of your life, you're much more likely to succeed.

Success in science is a combination of really hard work and persistence there's

some luck involved but the more effort you put into it the better your luck gets
