Youtube daily report w Nov 22 2017

Hi I'm Erin.

Every bite of food that ends up on your table has a story behind it.

A majority of the time you as the consumer never have the opportunity to hear that tale.

Today follow the journey of one loaf bread from our kitchen into our customer's hands

on Our Wyoming Life.

It's almost Thanksgiving and it's time for our first winter market.

We now have a new opportunity to sell our products year round.

We also have an opportunity to change some of the things that we are producing to fit

the seasons.

Apple Praline Bread screams fall and Thanksgiving and it looks divine.

This is a new recipe.

Something we have never made and sold before.

The journey of bread started 2 months ago.

In Cincinnatti.

Odd right?

Back in September Mike traveled to Cincinnati to a YouTube creators conference.

While he was there he met Kristen from Six Sisters Stuff.

You might have heard about Six Sister's Stuff.

They are dominating the cooking world.

They have cook books, blogs, a huge Facebook following and an incredible YouTube Channel.

As everyone got to know each other and learn about each other's channels Mike and Kristen

talked about how baking is a part of what we do that we haven't had the time to show

our audience yet.

An idea was born.

To take a recipe from Texas, to Cincinnati, to Wyoming to our customers plate.

Once Mike told me about this idea I was thrilled!

I immediately dove into their YouTube channel and started watching all of their videos.

Once I binged myself on YouTube I immersed myself in their blog and I found the holy

grail I was looking for, Apple Praline Bread.

When I saw it I knew that this was a must bake for me.

Kristen and I started chatting through email and sharing our baking stories.

Now market day is almost here and it's time to bake!

We start the baking by preparing the apples.

Several different varieties, some sweet some tart are peeled and finely chopped.

The recipe is being doubled so 3 cups are needed.

Next all the dry ingredients are measured and sifted into a bowl.

Wet ingredients are then added to a mixer.

Sour cream, sugar, eggs and vanilla are mixed on low speed until they are thoroughly blended.

Dry ingredients are added in slowly.

Once everything is incorporated it's time to fold in the apples.

Chopped pecans also get folded into the batter.

Mini 1 pound loaves are used for market.

An ice cream scoop making scooping the batter easy and helps keep the bread loaves consistent

in size.

Once all the batter is scooped there are 9 mini loaves.

More chopped pecans are sprinkled on top and lightly pressed into the batter.

Everything is now ready for the oven.

The oven has been turned on and preheated.

Baking for market is always done on the convection setting.

It helps the multiple loaves on multiple racks bake evenly.

Once the loaves look golden brown and delicious a toothpick is used to test for doneness.

It comes out clean and the bread is ready to come out of the oven.

It's placed on a cooling rack where it will rest for about 15 minutes and then removed

from the pan to cool completely.

Later that evening after all the baking is done and dinner has been eaten and a little

birthday celebration has happened for Mike it's time to make the glaze.

It's simple.

Brown sugar and butter is all that's needed.

It's warmed in a sauce pan until it boils for one minute.

Make sure to stir constantly until the mixture is thick and delicious.

Clear clamshells are used for packaging the bread.

This will contain and glaze that runs down the side and avoid any sticky hard to unwrap

packaging for market.

With the loaves in the clamshells the glaze can be placed on top of the bread.

It looks amazing and the glaze taste amazing.

Our customers are going to love this!

After all of our baked goods are packaged everything can be placed in a large tote and

is ready for transport to market.

The next day at market everything gets hauled in and placed on our tables and we are ready

to sell our products to our customers.

The Apple Praline Bread was quick to sell and before we even left market we received

an amazing review on Facebook.

It's always nice to hear that our customers will want to purchase the item again.

Apple Praline Bread might just become a staple of our market baking rotation!

Optogenetics: Principles and approaches - Duration: 4:45.

Optogenetics - a technique which combines genetics with light stimulation

is starting to be used in cell and developmental biology to control basic biological processes.

For example we can specifically guide the movement of a cell in a certain direction.

We can regulate cell signalling pathways by controlling the activity of membrane receptors

or downstream effector proteins.

We can also induce and prevent acto-myosin contractility of a cell or tissue.

We can control life death decisions, such as triggering apoptosis upon light exposure.

And finally, we can enforce a specific cell fate by the light-dependent control of cell differentiation.

Optogenetic techniques have been implemented in a wide range of organisms

and here are just a few examples of these ...

The biggest advantage of optogenetics compared with classical approaches for controlling biological processes

is the spatial-temporal precision it can achieve.

Since we use light to trigger the system, we can for example specifically activate individual cells in a tissue.

In certain experimental setups we can even achieve subcellular precision,

such as the specific induction of cell protrusions at defined sites.

Just as important as the spatial precision, is the fact that optogenetics enables us to

activate a system at precise time points to investigate dynamic biological systems.

By changing the frequency, amplitude and duration of the activation pattern,

it is also possible to directly modulate the kinetics of dynamic biological systems.

On the molecular level there are currently three major groups of optogenetic systems.

The first known system was the channelrhodopsin-like photosensitive ion channel.

This channel opens in response to light and facilitates a flux of ions to change the membrane voltage potential of a neuronal cell.

Photosensitive ion channels are widely used in neurobiology and approaches based on this system

are referred to as 'neuronal optogenetics'.

The second system involves proteins that undergo light-induced dimerisation.

Light exposure induces a conformational change in the photosensitive protein domain.

Then a specific interactor exclusively binds the photosensitive protein in its excited state.

Using this light-dependent heterodimerisation, a fused target protein can be recruited to the plasma membrane.

The third class of optogenetic systems is based on photocaging.

In the dark, the photosensitive protein of interest has an inactive conformation, usually because of allosteric autoinhibition.

Upon light exposure, intramolecular unfolding caused by minor conformational changes

activates the target protein or exposes dedicated signalling motifs.

Applications which make use of light-induced dimerisation and photocaging systems

in cell and developmental biology are referred to as non-neuronal optogenetics.

This e-learning course focuses exclusively on these more recently evolved systems

that can be applied in essentially all cell types and model organisms.

Optogenetics makes use of a defined set of molecular approaches.

The most commonly-used approach is the recruitment of a protein of interest,

tagged with a photosensitive domain to a specific anchor protein upon light activation.

Using the same principle, a protein of interest can also be sequestered away from its site of action.

Removing a protein from its natural localisation into a different cellular compartment essentially removes this protein's function.

Some photosensitive proteins tend to form big homomeric clusters as a result of light activation.

If the target protein is catalytically active when present in high density, then clustering results in

the formation of active signalling hubs inside the cell.

Optogenetics can also be used to control gene expression.

Transcription of a gene of interest can be regulated by making use of these kind of dimerisation strategies.

For example, the light-induced complementation of a split transcription factor

or recruitment of a transcriptional activator can trigger target gene expression.

Using a slightly different approach it is also possible to directly control the enzymatic activity

or binding affinity of a target protein.

A conformational change in the photosensitive domain, bioengineered in the protein of interest

can expose a hidden signalling motif or relieve the protein from allosteric autoinhibition

Average American vs Average Mexican - People Comparison - Duration: 7:18.

The nearly 2,000 mile (3,201 kilometer) United States border with Mexico is the most frequently

crossed border in the entire world.

According to the BBC, the border between Tijuana and San Diego is the busiest land border crossing

anywhere.

You might think this close proximity and the fact that people are continually moving between

the countries should mean the two nations have a lot in common, but that's not exactly

the case.

The U.S. probably has more in common with the UK, a country that lies over 4,000 miles

across the ocean.

Mexico's many indigenous civilizations were colonized by the Spanish in the 16th century,

while the USA was a colony of Great Britain.

This made all the difference.

And today we will discuss these many differences, in this episode of the Infographics Show,

The Average American vs. the Average Mexican.

First of all, let's have a quick look at the two countries our citizens call home.

The USA is the third (some sources say 4th) largest country in the world with a landmass

of 3,796,742 square miles (9,833,520 kilometers squared).

Today it has a population of 325,365,189 people, making it the third most populated country

on the planet.

Mexico has a land mass of 761,610 square miles (1,972,550 kilometers squared) making it the

13th largest country in the world.

The population is 129,669,477 people, making it the 10th most populated country in the

world.

So, who are these people?

The USA is said to be 72.4 percent white.

Other sources have put this estimate as high as 77 percent.

What we do know is that most of these white Americans identify as having ancestry from

England, Scotland, Ireland, Germany, France, Italy, Poland and the Netherlands, among many

other European countries.

The U.S. Census Bureau states that the next largest ethnic group is Hispanic or Latino,

at around 17.6 percent of the population, followed by black - 13.3 percent; Asian – 5.6

percent; and native American – 1.2 percent.

Multiracial, meaning people that identify as having mixed ancestry, is around 2.6 percent.

U.S. Census Bureau data also stated in 2016 that 5.6 million Mexican nationals were residing

in the U.S. without authorization.

As for Mexico, the largest ethnic group there identify as Mestizo.

Mestizo is a group – around 62 percent of the country – that are of mixed ethnicity

between the indigenous people of the country and Europeans.

This would include someone that has blood from Spanish colonists as well as Mexico's

indigenous peoples.

The next largest group at 27 percent of the population are Amerindians, which can draw

comparisons to Native American Indians.

Think of the Mayas from the Yucatan or Chiapas or the Náhuas, descendants of the Aztecs.

White (or European) Mexicans make up 9 percent of the population, while black Mexicans and

Arabic Mexicans are thought to make up about 1 percent each of the population.

With mixed ethnicity comes mixed religions, and the USA has quite a few on the go.

Nonetheless, the USA is said to be the world's biggest Christian population, many of whom

are still quite devout compared to their European counterparts.

More than 70 percent of Americans say they are Christian, and the next largest group

at around 23 percent aren't really anything at all.

Much smaller groups of people are Muslim, Jewish, Hindu and Buddhist.

Mexico is quite similar in this respect, with 93% percent of the population identifying

as Christian.

The only difference is that 82.7 percent say they are Catholic, while other Christian denominations

make up around 12 percent.

It's thought that only 20.8% of Americans are Catholics.

About 4.7 percent of Mexicans have taken the "don't believe" or "don't know what

to think" route, while Judaism, Islam, Buddhism, and native religions make up a very small

percentage.

Mexicans predominantly speak Spanish, but some people are bilingual or even trilingual,

speaking also English and/or a native language.

The vast majority of Americans are English speakers, although almost 40 million people

say they're mother tongue is Spanish.

Smaller groups state that they are Chinese, French, Tagalog, Vietnamese, Korean and German

speakers, in that order in terms of numbers.

What about the demographics of these people?

Well, the median age in the USA is currently 37.8 years old.

Life expectancy in the U.S. is currently 79.8 for both sexes, which is the 42nd highest

in the world.

The highest is Monaco, at the ripe old age of 89.5.

Average life expectancy in Mexico is 75.9, the 93rd highest in the world.

The median age in Mexico is 27.9 years old.

As for what we look like, the average American man is 5 feet 9 or 9 and a half inches (178.8

or 179.8 cm).

He weighs 195.5 pounds (88.6 kilos), according to the Center for Disease Control.

The average American woman according to the same source is five feet four inches (164.5

cm) and she weighs on average 168.5 pounds (76.5 kilos).

The average Mexican man stands at 5 feet 5 1⁄2 inches (167 cm) and weighs around 165

pounds (74.8 kilos).

We must point out here that sources are all over the place on this average.

The average Mexican woman stands at around 5 feet 2 inches (158.4 cm) and weighs 151

pounds (68.4 kilos).

Both countries are considered fat.

According to the World Health Organization in 2017, 33 percent of American adults were

obese and in Mexico 32.1 percent of adults were obese.

It's seems one thing these nations have in common is a love of over-eating.

As for how we live, we know that the USA is a far richer country and we know from all

our other shows on the topic that the USA has the most billionaires in the world.

But what about the average Joe?

The median annual individual income in the U.S. with both men and women taken into account

for a 40 hour work week is $44,148 per year, according to a 2017 Bureau of Labor statistics

report.

Mexico's median income was closer to 12,000 dollars per year.

Mexico may have 15 billionaires (some sources say 16) but the country has widespread poverty.

A 2015 report stated that 20% of Mexicans are worth no more than 80 dollars, with half

of the population living in poverty.

That 20 percent, it was reported, don't have the money to eat three meals a day.

The same report stated that, "From 34 member countries of the Organization of Economic

Cooperation and Development, the gap between wages and hours worked is larger in Mexico

than in any other member country."

To rub salt in the wounds, according to OECD, that year Mexicans worked on average 2,327

hours, while Americans worked 1,796 hours.

The United States is seen as a very hardworking country, too, in terms of hours worked and

holidays taken.

According to the Center for Poverty Research in 2015, the official poverty rate in the

U.S. was 13.5 percent.

Financially, Mexicans seem to have it much harder, but what about happiness levels?

According to the World Happiness Index in 2016, which takes into account money, work,

lifestyle, safety, freedom, etc., Denmark is the place to be.

The USA came 13th on the list and Mexico came 21st on the list.

That isn't bad, considering that Mexico ranked higher than the UK.

In 2017, though, Mexico and the UK basically changed places and United States dropped one

place.

In spite of low wages and some parts of Mexico being notoriously dangerous, it does seem

that "subjective well-being" is pretty good.

And that's what counts, right?

In fact, in 2014, a Pew research study found that Mexicans were the happiest people of

the 43 participating countries, which included Germany, France, South Korea, Japan, the US,

and the UK.

One Mexican news media outlet wrote a story with the headline, "Mexicans: fat and happy,

hard-working."

People have actually addressed this phenomenon and said one factor is that Mexicans have

much stronger family ties, but also live a more active public life.

While we hear a lot of negative news about Mexico, we don't generally hear about the

warm climate, community spirit, rich history, beautiful beaches; the streets often full

of music and celebrations, the general daily joie de vivre.

Kellyanne Conways Admits Trump Won't Denounce Roy Moore Because "We Want The Votes" - Duration: 3:57.

For those of you who've been paying attention, you should know by now that Donald Trump has

been eerily silent about the entire Roy Moore pedophile fiasco going on down in the state

of Alabama.

Well, as it turns out, Kellyanne Conway went on Fox and Friends recently to explain why

the President doesn't want to weigh in on the fact that the Republicans senate candidate

has been accused by several women of attempt to date them while they were in their teens

and he was in their 30s.

According to Kellyanne Conway, the reason Donald Trump is being so quiet is because

we need the votes.

What

she's saying is that Donald Trump doesn't want to screw up the campaign because they

need Roy Moore's vote in the senate in order to pass his disastrous tax-cut package that's

going to hand everything over to wealthy elite.

Basically, to boil this down to its most basic components here, Kellyanne Conway, White House

counselor, says that the President won't speak out against potential pedophilia because he

wants to give wealthy people, like himself, a tax cut.

That is 100% what all of this is all about.

That is why the President will not attack Roy Moore.

This is why he will not go after this man's ethics.

To be honest, I'm sure a large part of it also has to do with the fact that Donald Trump

has been accused by 16 women of improper sexual advances, sexual harassment and in some cases

actual sexual assault.

Getting involved in the Roy Moore thing is probably going to bring those issues back

up to the surface.

However, that has not stopped the President from weighing in on Al Franken, calling him

Al Frankenstein and attacking him for the women who are now coming out against Al Franken.

The bottom line is this: In Republican world, pedophilia is totally cool.

Being a hypocrite is perfectly fine.

As long as you're willing to go to bat and give wealthy American citizens everything

they want, Republicans will overlook any of your past sins.

That is exactly what Kellyanne Conway was describing to Fox and Friends earlier this

week during her appearance.

That is why Donald Trump will not speak out against Roy Moore.

How Disney Rides Know Our Names - Duration: 4:07.

Subscriber Dante Rodriguez asks "As the boat my Father and I were riding in approached

the end scene of it's a small world, the screen changed from goodbye in a foreign language

to saying goodbye to my father and I.

What struck me as strange was that we went through the normal queue.

We never actually scanned our Fastpass at any point while getting on the ride.

So I was hoping you could shed some light as to how it knew our names."

That's a great question Dante!

To anyone who doesn't know what Dante is referring to, the introduction of Magic Bands

in 2013 changed the way guests visited Walt Disney World.

Today with a trip to the resort, guests are given wristbands, called MagicBands, that

they can use in multiple ways.

They act as tickets to the parks, keys to your hotel room, and a way to pay for food

and merchandise.

Perhaps most relevant to this question however, the MagicBands are also used when getting

onto rides with a FastPass+ reservation.

Guests scan their band at a small stand at the entrance of the attraction to confirm

their reservation and then walk onto the queue.

However if you're just getting on the normal standby queue, you never have to scan anything.

So to Dante's question, if he and his father didn't scan their bands at any point in

getting on the ride, how did the ride know their names at the end?

Well the answer to your question Dante is that while you may have not scanned your MagicBand

on it's a small world, the ride scanned you.

You see, MagicBands actually come with two antennas built into it, a close range RFID and a longer

range ultra-high frequency radio.

The RFID, which stands for radio frequency identification, is the one we think about

when we think of the MagicBand.

It's the technology that allows us to tap our MagicBand to a FastPass kiosk or at a

cash register when paying for our lunch.

While the tech behind RFID would allow it to work at longer distances, it wouldn't

make much sense in this case.

After all, we don't want to accidentally pay for another person's meal or check-into

our Tower of Terror fastpass reservation before we mean do.

We definitely don't want to unlock our hotel room door without actually being at the door

itself.

The whole Mickey to Mickey design ensures that when we take these actions, it's because

we purposefully mean to.

That second half of the MagicBand technology, the ultra-high frequency radio, that's where

the Disney "magic" happens.

That's the part of the band that can be used from afar, meaning that a sensor you

don't see or even know about can read it and use the data to enhance your ride experience.

It's a number of hidden sensors in Be Our Guest that are used to triangulate your position

and let servers know what table you're sitting at so they can bring you the food you pre-ordered online.

It's a sensor in the queue of Rock n Roller Coaster that allows the digital concert posters

to update with guests' names and hometowns.

It's a hidden sensor in Frozen Ever After that scans your band to ensure that your ride

photo is linked to your PhotoPass account since there isn't a kiosk at the end of the ride.

And yes, it's a hidden sensor at the end of it's a small world that allows the ride

to customize those goodbye signs with the names of guests.

All in all the technology has some exciting possibilities.

We've already begun to see it used more and more in attractions as a way of adding

a personal touch.

We're also going to see a whole new use for it with the opening of Star Wars: Galaxies

Edge, as decisions made in attractions will carry over into how you're seen throughout

the rest of the land.

So like most other instances, the next time you're at Disney and feel wow'ed by that

Disney magic, keep in mind that there's some pretty clever technology at work.

The Untold Truth Of Celine Dion's Sons - Duration: 4:09.

Despite an iconic music career, Grammy-winning singer Celine Dion says being a mother is

her proudest achievement.

Following the death of her husband Rene in 2016, Dion's tightknit bond with her kids

has been increasingly in the spotlight.

Here are a few facts about the boys you may not have heard yet.

IVF journey

All three of Dion's sons—Rene-Charles and twins Nelson and Eddy—were conceived utilizing

in vitro fertilization.

Dion has been open about her struggle to conceive.

She received six IVF treatments before conceiving her twin boys in 2011.

One of those IVF cycles resulted in a miscarriage.

Just days before her twin boys were born, Dion also revealed in an interview with Quebec

TV that she was originally pregnant with triplets.

She told the Daily Mail,

"With any pregnancy, whether it's through IVF or not, you feel a danger.

You have to remain positive and try to relax as much as possible.

I always say that my children's first country is inside of me, so I try to make it a good

one and be healthy."

Name game

Speaking to Oprah Winfrey in 2011, Dion admitted she and husband René Angélil initially had

a tough time picking out names for their twin boys, born October 23rd, 2010.

In fact, at the hospital, they were simply known as Baby A and Baby B. But a week later,

the couple came to a compromise.

Dion told Oprah,

"The first thing that popped in my heart was Nelson because we had the privilege of meeting

with Mr. Nelson Mandela."

Eddy was named after another one of the couple's heroes: Eddy Marnay, who wrote Dion's first

five French albums.

Touching tribute

Rene-Charles—who goes by the nickname "RC"—made headlines in 2016 when he gave a touching

and emotional eulogy at the funeral for his father in Montreal.

It had many heartbreaking lines, including, "You are a tough act to follow, but with your

help, everything's gonna be fine.

Dad, I promise you that we're all going to live up to your standards."

Angelil, who worked as a music producer and also served as Dion's manager, died on January

14th, 2016, after a long battle with cancer.

In the wake of her husband's death, Dion admitted she struggled to find a way to explain the

loss to her twin boys.

She found a gentle way to explain the situation by comparing it to one of her kids' favorite

Pixar movies ...

"Do you remember the movie Up?

It's one of our favorites."

"I said, 'Well, yeah.

Y'know, today, Papa went 'up.'"

Dion told People magazine the comparison is the thing she's "most proud of" regarding

how she broke the news to her boys.

"I did not want to relate the passing to doctors and have them be afraid."

"Up is a good thing.

Up is uplifting.

We're going to call this — We're going to celebrate right now because Papa is no more

in pain.'"

Tightknit twins

The death of Angelil has brought Dion closer to her twin boys.

In fact, Dion told the Sun in April 2017 that they all sleep in the same bed, saying,

"I organize myself to not feel lonely.

So I got myself a huge, huge, huge bed and I sleep with my twins.

[...] They are comforting me a lot.

I need them.

I need them close."

The situation, of course, remains temporary.

"When it's time for them to say they want their room, their room's ready."

Stand-up teen

Dion's hectic life as a working mother has its many stressful moments, but one thing

she doesn't have to worry about is dealing with an angsty, rebellious teenager at home.

She told Us Weekly in 2015 that RC is "a wonderful young man."

Like any good Canadian, RC has developed a love for playing hockey … and Dion has become

the most incredible hockey mom of all time.

In August 2017, she was videotaped dancing and clapping in the stands during one of RC's

hockey games near Montreal, where she also took the time to take selfies with fans.

Speaking to Extra in February 2017, Dion revealed that after Angelil died, she wanted to make

sure 16-year-old RC never felt like he had to fill his late father's shoes ...

"And I said to him that I never want him to feel the pressure that he's the man of the

house.

'No, you have your own dreams."

"Your father is not here but he's in your heart.

He's always going to be supporting you in a different way."

Hey everybody out there!

Coach Dan Long with Get Lean In 12.

In today's short video, I'm going

to answer the question is it really

necessary to avoid sugary desserts, like what

you see I have here, to get the body that you desire.

Such as cookies, cinnamon rolls, carrot cake,

other kind of Chips Ahoy cookies that some of you I know

might be your favorite.

Well I'm going to answer the question today

if these should be avoided to get the body that you desire.

So stick with me here.

Now I want to explain who I am.

I'm actually Dan Long and I am part Get Lean In 12.

I'm one of the coaches here at Get Lean In 12.

I've been blessed to be a part of the industry for people

like yourself to help in the fitness and nutrition niche.

I went through a serious custody battle

in my time to be able to finally find my passion with what

I love to be able to do, which is fitness and nutrition, which

is why I'm here to share these amazing tips with you today.

is going to help you combat your favorite desserts such as what

you see here.

So stick with me and don't click that link just

yet, and listen to the tips that I have for you today.

It's going to be a 12-minute metabolic protocol that I'm

going to give you that's going to help you keep

the body that you desire by eating

these desserts with the tips and the ways

that I actually give you right now.

I do want to let you know that this system

that I'm going to introduce you to,

once I give you all these tips, it helped my wife--

over 40 mom of three children--

combat Crohn's disease.

She has an autoimmune disease and, as anyone knows,

Crohn's it can kill you.

Stress is one of the biggest factors when

it comes to Crohn's So with this protocol

I'm going introduce you to here shortly she's

able to actually stay in remission,

and it's been an amazing discovery for us

that I want to share with you today.

So let's get into it.

Can you have these desserts and have the body that you desire?

You're ready for this?

In big, bold letters--

yes!

You can have these desserts if you use these tips that I'm

going to give you, strategically, to get

the body that you desire.

With these tips-- let's do tip number one.

Tip number one, do not plan to devour a ton of this all at one

time.

Do not eat until you're uncomfortable.

That is a major key to the success

of being able to eat these type of foods

and still get the body that you desire.

So let's go over that one more time.

Do not plan, meaning don't go get

a bunch of these cookies or numerous different cakes--

or I know you can see this cookie here, this is delicious,

these are amazing.

Then we also have this cinnamon roll.

Don't plan this.

So what I mean by that is when you go to the grocery store

don't over indulge by getting too many items.

All it's going to do is stare at you in the face

and you're going to want to eat all of it.

So do not plan, and if you do consume these do not over eat.

That is key.

Tip number two, if you do consume

this what you need to do is you need to go to low carb

right afterwards for at least two to three days.

So what do I mean by low carb?

Low carb is just the normal amount of fat you can have,

which is the fingertip.

Your thumb right here.

Carbs, normally, you would eat a fistful.

That's normal carbs.

Low carb would be your fat and then

you can consume the palm of your hand of protein--

and remember protein burns fat.

So if you consume the protein with the fat

and you go low carb the glycogen, and your insulin,

that you actually just spiked by having these desserts

will put you into a fat burning environment.

So that's number two.

Tip number three, timing is everything.

You're probably wondering when should I eat these desserts?

Obviously, you don't want to eat this before you go to bed.

You want to do that at least four hours before you

go to bed.

Timing is key when you want to use this to maximize

the fat burning environment.

What you need to do is time this where

it's right after exercise.

I know that sounds crazy.

Cortisol levels are going to put you at the maximum fat burning

environment right after you do an exercise session that's

intense exercise.

The 12-minute metabolic protocol--

if you do that your cortisol levels now

within a short period of time, meaning I

would probably not have any of these dessert at least

no more than 1.5 hours--

1 and 1/2 hours--

after your intense exercise.

That will help maximize the sugars--

and, again, do not overeat.

Do not overindulge.

This is a very large cookie.

If you have one of these cookies I'm

sure you're probably going to get full.

Now for a guy my size I'll probably eat a couple of them.

But it also gives me the craving that I'm desiring

and it keeps me from overeating and or cheating

on my normal diet.

Carrot cake, obviously, you don't need two pieces.

A cinnamon roll-- have a cinnamon roll.

Enjoy it.

Timing is the key.

After exercise feel free to indulge in one

of these amazing desserts.

But, remember, don't do it later than 1.5 hours

after your high intensity exercise.

Then for two to three days go low carb,

and then you can do that again.

Just don't do it back-to-back days.

Don't do it two times in one day.

Just do it once.

Get your cravings out and I promise you

the fat burning environment will be at an all time high.

Now tip number four, the exercise that you do cannot

just be some steady state cardio.

It needs to be high intensity that's going

to get the adrenaline flowing.

It can be 12 minutes and it can be metabolic.

And when you do that your cortisol levels are

going to be at the maximum level to burn fat.

That is when you want to consume desserts like these.

I don't care what it is that you love in desserts.

You can have anything you want.

Just don't overindulge, don't plan.

Remember what I said as far as intensely.

You got to have intense exercise,

and you also need to make sure that timing

is done within 1.5 hours.

And you should be four hours before bed time consuming

these desserts.

Now because of that high intensity exercise,

what I just told you to do, I have something specifically

designed for the hormonal condition for people

in their 40s, 50s, and 60s here today

that I want to introduce you to.

It has age specific components that are in it that are

specifically designed for people that are over the age of-- it

could be 35--

but specifically over 40, like myself.

I'm 44-years-old and I tell you I feel 25.

Only because I live the life of exactly what I'm giving you

here with these helpful tips to help you maximize and have

the body that you desire.

The only way it's going to happen,

though, is to follow that exact protocol.

Now that protocol I just so happen to have here today.

And included in that protocol is, also,

a program that's called Lean19.

Exploring different optogenetic systems: Photocaging - Duration: 3:26.

Photocaging is an optogenetic method that allows us to directly regulate the function of a protein

by tagging it with a photosensitive module.

The most commonly used class of photoreceptor modules for photocaging are the LOV domains.

LOV stands for light, oxygen or voltage.

The LOV core domain consists of a classical sensor domain called PAS, which is made up of

a five stranded antiparallel beta sheet and alpha helical connector elements.

Once exposed to blue light, one of these helical elements undergoes a conformational change.

This can affect protein activity by interfering with allosteric regulation or by unmasking an active site

One of the most widely used LOV domains is LOV2.

Like other LOV domains the PAS fold non-covalently binds to a flavin chromophore in the dark.

When excited by blue light, a covalent bond forms inside the LOV domain that is transmitted to the J-alpha element,

causing a conformational change of the LOV2 to domain.

LOV domain-containing proteins are found in many organisms

including bacteria, fungi, algae and higher plants.

Each LOV domain has unique properties in terms of activation kinetics, sensitivity and relaxation time.

And these should be carefully considered when developing your optogenetic system.

Photocaging has been used to control enzymatic activity and protein-protein interaction.

The target protein is functionally trapped in the dark, but once exposed to blue light,

the conformational change in the LOV domain results in activation of the target protein.

Photocaging can also be used to unmask a short signalling motif.

In this case, the J-alpha helix of a LOV2 domain is engineered to contain a sequence motif that is hidden in the dark.

When exposed to blue light, the helix unfolds and presents the sequence motif,

enabling other proteins to bind to the target protein.

Mask signalling motifs can include:

trafficking motifs, recognition motifs for post-translational modifications

or protein-protein interaction motifs.

Using this approach we can also shuttle a target protein between different cellular compartments

In this example, the hidden trafficking motif is a nuclear export signal,

and the protein of interest localises to the nucleus in the dark.

Following blue light illumination, the nuclear export signal becomes exposed

forcing the protein into the cytoplasm.

One limitation of photoaging is that it's time-consuming,

it usually requires a screening of different LOV domains, insertion positions within the target protein,

and lengthy optimisation protocols.

Photocaging might not work for every protein of interest but it does provide a very elegant and powerful

approach to study protein activity.

HAA recently reached 800 students, which is a huge milestone, and with that, we thought

that we would roll out some new social media initiatives to help keep you informed and

answer any questions that you might have about HAA.

We're going to be doing that in a couple of ways: I would like to remind you to always

to keep you updated -but now, we are going to be rolling out a couple of different YouTube

series that will hopefully help you and your family stay happy here at HAA.

One series that you'll see starting at the beginning of December, and continuing into

2018, is going to be our "Month In Review" series.

At the beginning of each month, we will be posting a video that will tell you a little

bit about what has happened at HAA during the previous month, and what you can look

forward to for the future.

In those vlog style videos, I'll be showing you a little bit about what it's like to be

here on a daily basis at HAA, as well as answering any questions that you may have or giving
