2-Minute Neuroscience: Opioids - Duration: 2:01.
Welcome to 2 minute neuroscience, where I explain neuroscience topics in 2 minutes or
less.
In this installment I will discuss opioids.
The term "opioid" refers to substances that bind to opioid receptors, usually to
exert pain-relieving effects.
Opioids can be found naturally in the opium poppy, or they can be partially or completely
synthetic.
In addition to pain relief, opioids also cause sedation, anxiety reduction, and euphoria,
along with a variety of other effects.
When opioids bind to opioid receptors, they promote potassium conductance, which makes
a neuron less likely to fire an action potential, and they inhibit calcium conductance, which
makes a neuron less likely to release neurotransmitters.
What effect these actions have depends on where the receptors are found.
When opioid receptors on neurons in the spinal cord that carry information about pain are
activated, it can inhibit the transmission of pain information to the brain.
Opioids can also act on descending pain modulatory systems to reduce pain.
For example, by binding to receptors an area of the brainstem called the periaqeuductal
gray, opioids can prompt the inhibition of pain signaling in the spinal cord via a pathway
that first synapses in the medulla.
Opioids can reduce the emotional impact of pain by acting in brain regions like the anterior
cingulate cortex, and they increase dopamine levels in areas like the nucleus accumbens,
which may contribute to the reinforcing qualities of the drugs.
Exposure to opioid drugs leads to tolerance, which involves a decreased response to the
drug and a need to take more to produce the same effect.
The mechanisms of opioid tolerance aren't fully understood, but they're thought to
involve adaptive changes like a reduction in the number of functional receptors for
the drug to act at.
Adaptations in response to the presence of the drug also lead to dysregulation when the
drug is removed, and withdrawal effects that are opposite in many ways to the typical effects
of the drug.

I'm in the car, we had breakfast and we got ready. Today is my last work day in the kids their holiday.
Next week school is starting again for the kids. So the summer holidays are finished and it's my last workday.
It's Thursday but tommorow it's Australia Day, so I'm off tomorrow.
So that why I wanted to bring the kids somewhere and we're going to Wildlife Sanctuary
For example.. they keep Koalas and Kangaroo's.. and all kind of other animals!
So that's where we are going now, it's a 30 minute drive
Carla can often choose the music in the car.. Which is what she is asking me here! ;)
It's 12.30 now and we are having something to eat.
We'll stay here after lunch as well, it's really fun for kids here!
I just held a lizard... *sos*
It's something I would never have done.. even though I held a snake once.
But they suddenly came to me here and the man said he would give the lizards to the adults so they could decide themselves if their kids could hold them or not.
Well.. he started with me and I thought: 'No way that I will hold that thing myself.'
But I didn't want to say that when the kids where next to me. :') #gottalooklikeacoolaupair
So I held a lizard and the kids as well, they had a snake as well I didn't held that.

Quite funny, anyway it's really nice for kids here!
In the meantime we left the park and went to the beach!
And we're back home! And I look really burnt.. - Maybe I am.
We've been back for a while.. It's 6.15 and the parents just left the house.
The mom has a party and is staying with a friend afterwards and the dad is going to Japan for 10 days.
So tonight I'm alone with the kids.. I'm going to make dinner now, pasta; because it's simple and quick.
And than I'm bringing them to bed. But I don't want to bring them to bed too early because usually they go around nine.
And than they sleep long as welll. And when I bring them to bed early; they wake up earlier as well
And I sleep next to the kids.. so sometimes they go to my room in the morning.
And tomorrow I'm off.. :')
Anyway, I'm going to make dinner first; pasta like I said. Super simple, tortellini with spinach and ricotta..
With this sauce.. actually I only have to heat this up. But there isn't much more at home, so simple and easy!
This happens so often.. That Carla and I laugh about something.
Thomas has no idea what is happening but he still laughs with us, because he sees other people are laughing.
It's still quite early.. but the kids where tired already and they were just haning around..
But I don't want to bring them to bed yet.. So we're watching the serie Tarzan now!
I wanted to show them the film.. but it's not on Netflix so that's why we're watching the serie instead.
And after that I will bring them to bed!
It's ten past 9 and the kids are in bed! Which is really on-time compared to there usual time!
And they went to sleep super quick.. Or wel.. they're not sleeping yet, they're still talking
But they were okay with the fact that I would leave really quick.
I even got a teddybear from both of them to sleep with, so I wouldn't be alone either. Super cute!
So yeah, they're in bed! I hope today was a bit clear, the parents weren't there
Only before dinner the dad was there for a bit and the mom wil be back in the morning.
So it's the first time I'm alone for the night.. And usually the kids sleep through the whole night!
But there is still something inside me what says: "What if they wake up tonight?! When you're alone with the kids."
Anyway, probably that will not happen.
I really liked today.. I am super tired, I think u can tell.

For VOA Learning English, this is Everyday Grammar.
This week's Everyday Grammar looks at how to use the modals may, must and might.
Modal verbs (called modals for short) are auxiliary verbs that express a speaker's
attitude and the strength of that attitude.
There are about 17 modals in English.
They have multiple meanings and sometimes overlap in ways that are confusing to English
learners.
Today we will look at how we use these modals to express how certain, or sure, you are of
something.
Grammar expert Betty Azar explains that these modals tell us how sure speakers are about
what they are saying.
A person who is 100 percent sure uses the verb be, as in, "I am sick."
If they are mostly sure, say 95 percent, they will use the modal must, as in, "I must be
sick."
When speakers are about 50 percent sure, they will use the modals may, might, or could;
as in "I may be sick.
I might be sick.
I could be sick."
Might is used as the past form of may.
For example: "I may take more pictures."
(This is a direct quote) She said she might take more pictures.
(This is reported speech) Notice how may changed to might.
Modals change to a past form in reported speech.
Yesterday we had a staff meeting.
I looked around the room and noticed my co-worker Anna was not there.
I asked, "Where's Anna?" and got three answers from my co-workers.
Jonathan answered, "Oh, she may be making a video in the studio."
Kelly said, "She might have stayed home today."
Adam told us, "She called me to say she was doing an interview at the Capitol this morning.
So she must still be working over there."
In this conversation, you can see a change from may to might.
May shows the speaker is not sure in the present moment: "She may be making a video."
May changes to might to express a possible state in the past: "She might have stayed
home."
Finally, must expresses a strong certainty: "She must be working there."
People today do not always follow these rules about present and past tense for mayand might.
You will hear both words to express the same degree of certainty.
English speakers still express strong certainty in phrases like, "It must be love."
Listen for the word might in this song by The Cars.
You might think it's foolish Or maybe it's untrue
You might think I'm crazy But all I want is you
By using might, the singer is expressing about 50 percent certainty.
Now let's look at how we express certainty about the future.
My friend Andy has a test next week.
He has studied very hard for months.
I told him, "You will do well on the test.
Don't worry."
I believe with 100 percent certainty that Andy will pass the test.
On the other hand, Carrie, who has to take the same test, just began studying last week.
I warned her, "You might not do well on the test.
You should study more this weekend."
I am not so sure that Carrie will pass.
In fact, I doubt it.
I express that future possibility with might.
The idea of possible future events lets English speakers use may to talk about hopes.
You will see may on greeting cards and in prayers or religious writings.
A quick look at Google Ngrams shows that few people are using may in this way.
Now, it is much more common to hear "I hope that."
Traditional poems and prayers also use may to express positive sentiments.
This is part of an old Irish blessing: May the road rise up to meet you.
May the wind always be at your back.
May the sun shine warm upon your face.
With that in mind, the Everyday Grammar team says, "May you find our articles useful."

So when I was a kid, like most people my age, I was obsessed with Harry Potter
You know, I went to the screenings, there was a Harry Potter day camp I went to
I dressed up like Hermione for Halloween
as an adult im still pretty obsessed with Harry Potter
Where my Hufflepuffs at?! EY!
Some of my fondest memories as a kid
are being out on the school yard with all my friends
using coloured pencils as wands
pretending that the playground was Hogwarts
and going on magical wizard adventures in our imaginations
and the thing im going to talk about today is a lot like that
except the wands are better
and the magics better
and the castles
way better

Is not directly related to Harry Potter
It takes place in a universe that is very heavily inspired by
the universe of Harry Potter
but it is not a Harry Potter game
and a big part of that is youre not going to be playing a hufflepuff
you're not gonna be playing snape
because it takes place in this completely original world
you need to make up a completely original character
You're not going to be attending bothwell school of wizardry as yourself
Everyone goes as a character
Like a big 'ol adult game of make believe
When you sign up, you fill out this survey to let the organizers know
What kind of wizard you want to play
and what kind of wizard you definitely don't wanna play

Spotlight On: James Bond's Omega Watches.
The Omega Timepieces on 007's wrist.
Throughout the history of the James Bond franchise, the watches on Agent 007's wrist have been
as varied as the "Bond girls" on his side.
But that all changed with 1995's Goldeneye.
Coinciding with Pierce Brosnan's first appearance, was the introduction of a new Bond watch – the
Omega Seamaster.
Bond has been wearing theSeamaster since then, and with a 20-year long partnership, there
seems to be no looking back.
Here's a look at all the Omega watches that have graced the wrist of Bond, James Bond:
In 1995, Omega won the bid to be the official watch of Agent 007.
This was the start of the relationship between Bond and the Omega Seamaster.
The suave and sophisticated new James Bond, played by Pierce Brosnan, was the first to
strap on an Omega – and they chose a professionally-oriented diver watch.
Brosnan first donned the quartz edition Seamaster 300M Professional in GoldenEye, followed by
an automatic and chronometer rated model in the next three films.
The next decade saw a rougher, more modern James Bond played by Daniel Craig.
He still dons the emblematic blue Seamaster Professional at first, but he then switches
to the Planet Ocean.
Water resistant for up to 600M, it allows Bond to get into dangerous situations, even
underwater.
In 2006's Casino Royale, he wears the 45 millimeter, rubber strap Planet Ocean.
The watch proves pivotal in the movie as it helps save him from underwater trouble.
It was followed by two more Planet Ocean models, but the watch would no longer be used as a
gadget for the next two films.
In 2008's Quantum of Solace, he wears a smaller model with a 42mm case, metallic bezel,
and steel bracelet.
And for the 2012 movie, Skyfall, Omega released the limited edition Seamaster Planet Ocean
007 Skyfall Watch.
The watch Is a special version of the Planet Ocean steel model, outfitted with a matte
black ceramic bezel, and a 007 marker in place of the 7th hour.
Bond was most often seen wearing tool and dive watches, but he also donned an O-mega
Seamaster Aqua Terra in the 2012 movie, Skyfall.
This was a surprising choice, given that the Aqua Terra is simpler yet dressier than the watches
he used to wear.
He wore a nautical-inspired striped blue dial model, with a steel construction.
And finally, for the latest bond movie, Spectre, the Omega watch came back in a really big
way.
Not only did they create a specific limited-edition watch for the movie, it also returned to becoming
a gadget timepiece, after having no particular role in the previous 3 movies.
The Seamaster 300 Spectre LE features a vintage-military inspired look with its patinated indexes and
NATO strap.
It featured a countdown and a bomb that helped Bond save the day, once again.
The next Bond film is slated for release in 2019.
It's Daniel Craig's fifth, and final performance, as Agent 007.
We are psyched to see what he'll wear on his wrist.
And one thing is for sure - it will be an O-mega Seamaster.
