A drug's journey: how drugs get from the lab to the market - Duration: 3:21.

The story of a medication before it gets to the shelf of a chemist is a long one.

Less than 10% of drugs designed by pharmaceutical companies

actually end up making it onto the market.

That's because most don't pass all the hurdles on the way to being approved for sale.

Before a drug even comes near being tested on humans, it's tested on animals.

This first step helps scientists test the effects of the drug on vital organs and how

toxic the drug is at different doses.

Once this hurdle has been cleared, the drug must go through three phases of human trials

– known as clinical trials –

before it is approved for sale.

Phase one is the first time the effects of a new drug are studied in humans.

At this stage it's all about safety.

A small sample of up to around 80 healthy volunteers trial the drug to establish its

toxicity over a range of doses, based on the results of the animal studies.

If the drug is found to be safe, it enters the next phase of testing.

Phase two of the trial process focuses on the benefits of the drug:

whether the drug treats the target condition, or minimises its effects.

Several hundred patients with the condition are included in the trial sample, and if there

is evidence of a benefit to patients (with an acceptable level of side effects),

the drug progresses to the third phase.

Phase three is the most important phase of drug testing, and the last stage before drug

developers seek approval from regulatory agencies

– like the Therapeutic Goods Administration in Australia

or the Food and Drug Administration in the US – before it goes to market.

Phase three is where researchers seek the definitive answers on a drug's efficacy

and safety.

The number of participants involved, and the duration of the studies vary depending on

the product and target condition, but hundreds or even thousands of participants are often

involved.

And the best way of proving a drug's efficacy against the current standard of care is through

a randomised controlled trial.

Randomised controlled trials generally divide study participants randomly into two separate groups

- one group of participants receives the new drug,

while the other is a "control" group –

they receive either no treatment at all,

a placebo (which appears to be the treatment but has no active ingredient)

or the standard treatment available at the time of the trial.

The two groups are randomly allocated to make sure that effects shown in a trial are the

result of the drug itself rather than factors like age, lifestyle choices (like whether

the participants are smokers or live in a specific environment) or gender.

And to make sure that the effects aren't boosted or hampered by the patient knowing

which treatment they're getting, trials are generally what's called "blind".

Where possible, the researchers and treating doctors are also blinded, so no-one knows

what treatment a participant is receiving until the results are in.

Once the drug clears phase three, it generally gets approved for sale and hits the market.

But testing doesn't stop there; that's when phase four kicks in.

It's the final safety measure on a drug and focuses on its long-term effects and potential

use for treatment in other conditions or new populations, like children.

These studies generally involve a wide population receiving the drug (sometimes thousands) and

are often a condition of the drug receiving approval.

So when you take medication, you can be safe in knowing that it's gone through a process

of testing that can take up to a decade.

this game but I broke it down and it's going to be a very quick little

breakdown I actually had a lot more promise for Miami when I saw after you

know they had their bye week of course because the hurricane they get to play

Tampa Bay the first week and they have this sacrifice their bye week

Miami's getting a plus +11 going into Atlanta there's an over/under of 47 and

this 47 keeps changing on all different levels it's been at 47

seven and a half and I seen that 46 and a half same thing with the eleven that's

been bouncing back and forth on different I Vegas NFL lines against spread

lines and everything else so it's been tossed around a little bit but anyhow

break down if you've watched my other videos this one's not gonna be that

detail because there really wasn't much for me to break down I added up

everything that Miami's done I first I ended everything up with you know of

course being in London I tried to not look at that game as much but I did just

a little bit since you know I was twenty zero but they are international game and

everything else but anyhow Miami's average they give their opponents an

average of 16 point 75 per game they average right now scoring 10 points now

when they're away like going to Atlanta they average giving their opponents 18

and a half points and they average scoring 12 and a half points now the

difference is we got a Atlanta Falcons week five last week this is week six of

course they had a bye so now they're fully rested even if their players were

a little bit injured or sore they've got time to kick their feet up

and relax they averaged given their opponents 22 points and they've score an

average of 26 points now when they're at home since this game is at home

they average given their opponents 23 points how crazy is that Buffalo 23 they

give and Green Bay score 23 on them but at home they also score at least 25 and

a half points per game all right so I broke that down did the math

shuffle it all around you could see them on my other videos how I do it but when

I did it all bottom line it's not looking too

promising for Miami Dolphins this week and I really don't like these kind of

games when they're like this when I do all I expect

came to be 35 for Atlanta 13 for Miami with a total score of 48 that puts us

over 47 how I did that I came up with Miami's 13 I gave them their 13 I took

the odds I subtracted up all the math Atlanta 17 and a half game on there three

plus their Home field came up with 20 I added the 20 and the 13 together I

divided it by 2 I came up with 17 almost 16 75 then I added the 20 I came up with

37 I subtracted and then I added the 13 I came up with 50 then I did my tossing

turning her on picks and looking at everything

bottom line I don't like watching blowout games I don't think that many

people do either especially if they're fans for the Dolphins or if it was the

flip side I see this gave me an over 47 like I said 35:13 the only if yeah it

happens to be more like this alleged Falcons scored 32 points so let's

round it up 33 points it could be right under 47 if we have the 33 and let's say

Miami scores 10 then I make it 3 then you they score another field goal that

makes it 46 and we have it right under the point line I would not do anything

with this game and with the over unders unless you just want to have some fun

but I'll come 35:13 that's all I gotta say about this game sorry but I just not

going to be the it's not gonna be the greatest game let me know when your

She didn't specify if he was foreshadowing military action against North Korea or Iran.

She did make it known that the foreign policy focus is Iran and North Korea.

The US diplomatic pressure continues.

"I think we've got a lot of bad actors in the world, North Korea, Iran, there are

several examples there," Sanders said.

President Trump ignited a firestorm online Thursday night after he floated the possibility

of a "calm before the storm."

Mr. Trump, accompanied by First Lady Melania Trump and flanked by military members and

spouses for a photo op at the White House, said, "You guys know what this represents?

Maybe it's the calm before the storm."

Pressed on what he meant, Mr. Trump said "we have the world's great military people"

in the room.

Asked yet again for an answer, Mr. Trump said: "you'll find out."

The president reiterated those comments, almost word-for-word,

on Friday.

How To Make Creamy Pesto Pasta ~ Fettuccini with Basil Pesto Sauce Recipe - Duration: 3:32.

This pasta dish recipe is delicious and a definite comfort food. I'm Tess and

today I'm going to show you how to make a rich creamy pesto pasta. (intro music)

You can use any pasta that you like for this recipe but tonight I'm making some fettuccine.

Just cooking according to package instructions and saving a little bit of

the pasta water just in case I need to thin out my sauce. And just a reminder

that you'll be able to find this recipe and the list of ingredients in the show

more section below. Just click on the show more and it will open to reveal

lots of information. I've also included some online links where you can purchase

the ingredients and some of the equipment that I've used in making this

video recipe. If you have a chance please check it out. In a pan on medium heat I'm

adding in some butter, diced onions, mushrooms and lots of minced garlic.

Cooking and stirring for two to three minutes and then adding in some salt and

black pepper. Cooking and stirring for another minute or two and then removing

from the heat.

In another pan on medium heat I'm adding more butter and some flour. I'm making a

roux to help thicken the sauce. Just cooking and stirring for one to two minutes.

Now I'm adding in some heavy cream and turning up my heat to bring up

to a boil and then reducing to a simmer. Yep, heavy cream! I did say this was

comfort food! Adding in some salt and black pepper and a couple good

tablespoons of basil pesto. I'm also adding in some small broccoli florets.

Giving that a stir and cooking for two to three minutes.

I decided to also add a little heat with a pinch of cayenne pepper. Adding my

onion mushroom mixture back into the pan along with a handful of fresh chopped

parsley, Parmesan cheese and my fettuccine. Giving everything a good toss

to get incorporated. At this point if the sauce seems a little thick you can use a

little of the pasta water to thin the sauce.

You can serve this creamy pesto pasta as a main dish with a side salad and a

piece of bread or you can serve as a side dish. Tonight I'm serving with some

baked barbecue chicken and you can find my video recipe for the baked barbecued

chicken in the iCard above or in the show more section below.

I love the rich creaminess and the pesto really shines through. You can use the

vegetables of your choice and add in some cooked chicken or shrimp if you wish.

DOLLAR TREE CANADA HAUL! Sept 2017 - Duration: 10:43.

Hey guys, welcome back!

This is my first Dollar Tree haul of September.

I went to the Dollar Tree in Brampton to see if I could find some things that they did

not have at the Dollar Tree in Meadowvale.

Like I said when I went there in late August, they didn't have any Halloween things, they

didn't even have that much fall stuff, so I was surprised cuz I see all of you guys

hauling the Halloween stuff!

So the good thing is, I did find a bunch of Halloween stuff, as well as some things I

couldn't find - including this, which has been on my wishlist for months!

This camera tote bag - I had gotten the impression that they might not be selling it anymore,

and at the very last second while I was in the checkout, I saw it hanging there, and

I freaked out and grabbed one.

This one's kinda bent, but that's okay.

I just, I love this bag.

This is a bag I would actually use; that drawstring backpack that I picked up with this print,

I probably won't use, but that's okay cuz I have a DIY idea for it.

So I just picked up three food items today...

I picked up another pack of the hazelnut Happy Swing, because they're really really good.

The chocolate ones are pretty good too; I didn't like them initially because they're

not very sweet, but they kinda grew on me.

So I was going to pick up a second chocolate one as well, but they didn't have them there

for some reason.

And then I picked up these two bags of the Go(2)Snax chips and the cheese puffs, because

everyone has been saying they're really good, and I needed some chips!

I was very excited to find the chopping mats; I've seen a lot of people using them for crafting,

and some of them have actually been used IN crafts.

Actually I thought it was kinda funny, you can see on the wrapper - I don't know if you'll

see it on camera, but it says, you know, "Good for carving, chopping, and crafts"!

It's a two-pack, so for $1.25 you're getting two, so that's 75 cents a pop.

And I wanted to try it out as like a crafting mat, because crafting mats are really, really,

really, REALLY expensive, and I've heard that these work pretty well.

They look smooth, which is good, cuz I need a surface to use those Tim Holtz inks on...

because for those who don't know - I didn't know til I started trying to use them - when

you use the Tim Holtz, if your object is like, here - if this is what you're trying to ink

on - you have to start outside it and move your way in.

So you need a surface to do the inking on that is smooth, so it won't pick up too much

of the ink, or you're gonna have no ink on the blending tool by the time you get to what

you're actually trying to put ink on.

So this seems pretty smooth, so I'm gonna try it out, because again, the "real" craft

mats that are at Michaels are ridiculous; I think the cheapest one is like $32.00.

I got some Salon Selectives hairspray to go with that hair chalk I bought when I went

to Meadowvale, because I had no idea how to use hair chalk, so I was googling it, and

apparently if you want it to stay in your hair for longer than 30 seconds, you need

to like, hairspray it.

And again, like I've said before, I'm not good at being girly, so I have never owned

hairspray in my life, so I picked some of this up just to try with the hair chalk.

I did manage to find the inspirational calendar, so I was very excited to find that.

They definitely had more variety of calendars at this store, so I grabbed it.

This is probably the one I'm going to take to work, since it's all inspirational... and

again most of the prints are really big, so using it for crafting probably isn't going

to work.

So I will probably take this one to work.

I picked up this 3-pack of the three different sizes of the foam tape, a lot of people have

said that these work really well.

And then I just grabbed a 12-pack of different sizes of binder clips for like, holding stuff

in place while I'm making stuff.

I was going to use all the magnet clips that I have, but I'm thinking those might not hold

quite as tight, depending on what it is I need, so it doesn't hurt to have some of these around.

I've gotten a bunch of ideas for magnets and stuff lately, so I picked up two more sheets

of the adhesive magnet...

I already have one that I haven't used yet, but...

I have an idea!

And then I picked up some magnetic tape to try, and then the magnetic buttons for a crafting

idea that I have.

I forget who I saw haul this little butterfly box, but... actually I thought they got it

at Dollarama, and then I found it at Dollar Tree... but it gave me an idea for covering

it with this butterfly paper, so I picked up a second pack just to make sure I had enough,

because there's only two of the butterfly sheets in it, and I don't know if that's going

to cover this, so I picked up one more just to be safe.

I found a whole bunch of the Good Dog stuff, which made me VERY happy, cuz like I said,

they had almost none in Meadowvale.

So I just grabbed all the patterns that I really loved! [laughs]

I saw someone haul, I think the full-size notebook with this, "This Is How I Roll" pattern,

and that's the one I was looking for.

They didn't have a full-size so I just grabbed this, cuz it's... ridiculous, so [laughs]... I grabbed that...

I grabbed the one that says, "Goin' Out Easy Breezy", with the dog sticking it's head out

the car window...

And then I don't know if I've ever seen someone haul this pattern, but it's really cute.

It says, "Is it time now?

Is it time now?

How about now?" [laughs]

I thought it was so cute - it actually reminds me of my first dog.

He was a pointer, and he kinda had this pattern - like he was black here and black here and

had black ears.

So it actually looks a lot like my very first dog, so I kinda fell in love with that, so

I thought I'd grab those.

The last time I went to the Brampton store, they didn't have any stack packs; this time

they had a bunch.

I've seen this one before, the birthday paper...

I picked it up because I'm looking to do card-making, and obviously birthday cards are pretty popular.

And then I also picked up the blue Deco Paper, because I just really liked some

of the patterns, especially that one!

I'll do a quick flipthrough of each of them for you... but yeah, I just picked up each

of these cuz I thought that they were cute.

And they had the pink one too, it didn't really do it for me as much, and they had all the

other patterns of this pad I'd seen before...

I can't find the vintage stack anywhere and it's driving me crazy cuz that one looks gorgeous.

But I grabbed these two anyway, so I'll do a quick flipthrough of the patterns...

I only got two sets of non-Halloween stickers, although one of them I got for Halloween...

I bought these birthday hat & balloon stickers because I just randomly saw them sitting there

and again, I want to get into card-making; needed a couple more birthday things.

And then I picked up this sheet of the gem stickers to use as embellishments for some

of the Halloween stuff I want to make, so it's technically not Halloween, but I bought

it for Halloween.

And the last non-Halloween item, for crafting, that I bought was just this Chalk Writer,

because I got some chalkboard sheets in that destash I bought from CaliG here on YouTube

a while ago, and I didn't know what to do with them, cuz I don't have any chalk... so

I thought I would pick this up and see what I can do with them now that I have something

to write on them with!

And now we're getting into the Halloween stuff!

There wasn't too much there... they had two endcaps full of Halloween things, and they

had a sign up that said, "Pre-Season Halloween Sale"... not really sure what that means...

I assume that must mean they're going to have more Halloween stuff eventually?

I hope so!

I didn't find all of the stuff that I've seen people hauling, but I did find some stuff

- I don't know if I've seen anyone haul these stickers yet.

But I got two sets of each cuz they're all really adorable!

So I got two sets of this one that says, "Boo!", and it's a skeleton, and a little haunted

house, and "Spook Time", and a ghost wearing a witches' hat... so I got two of those.

I got this one that says, "Boo!", "Trick or Treat", "Scary", "Wow!", there's ghosts in

different hats, little owls, it says "Happy Halloween"... so I got two of those as well.

And this one with the witch, and another "Happy Halloween", a little mummy, and little token--

er, icons... and this person dressed as a ghost... so I got two of those.

And then lastly I got this one, with photo corners, I just noticed that now...

"Fright Night", "Party Time", "Spooky"... again it's just really cute.

Most of these are like pop-up stickers as well, and I got two sets of these also.

I hauled a bunch of the little felt stickers...

I just got one pack of the witches' hats and shoes, cuz they're not that cute... they're

cute, but they're not that cute.

But I did get two each of the other sets - I got the one with the pumpkins and the bats

and the ghosts, and the ghosts are kinda hidden in the back there... but I got two sets of

those...

And then I got two sets of the pumpkins, bats and cats... and I think there's something

else in there, more pumpkins?

More pumpkins.

So I got two sets of those as well, cuz they are really adorable, especially the cat!

I was very excited to find the LED lights!

I got two sets of the skulls... it's upside down... got two sets of those, and then I

got one each of the bats and the - whoops! - and the pumpkins, because I have a set of

bat lights and a set of pumpkin lights that I got at Target, back when Canada had Target.

I miss you, Target.

I miss you so much.

So since I already have two strings-- a string each of those, I thought I'd just get one more.

They don't quite match, but they're good enough.

And then two of the skulls, cuz I don't have any skull lights.

And finally I picked up some of the wraps and stuff.

I got this sparkle mesh; it's suggested to use for wreaths, but I'll probably just use

it for other things.

I got one with the silver spider webs and one with the gold spider webs... focus!

... cuz I just thought they were really cute.

I'm not sure if I've seen people haul these, but yeah, I just got one of each to play with.

And then I got three of the diamond wraps...

I thought there was supposed to be one with pumpkins on it, but I couldn't find it.

This is the one - it's probably not going to show, but this is the one with the spiders

on it... yeah, it doesnt want to show... it's got spiders on it, though! [laughs]

And then I got the purple one with the bats...

... and then I got the silver one with the skull and crossbones on it... again, just

to play with.

I hope I can find the pumpkin one, unless I'm hallucinating and there weren't any pumpkin

ones, but I could have sworn there were...

DOLLARAMA HAUL! Sept 2017 - Duration: 6:07.

Hey guys, welcome to my first Dollarama haul of September.

I just popped in when I was down at Dollar Tree in Brampton; I popped into the Dollarama

that was there to look at their Halloween stuff.

I did find a few things; this shouldn't be too long of a video, I would hope.

I picked up these little kids' skirts, um... augh... you can't see... Halloween skirts! They're $2.00 each.

I can't remember who I saw mention this - it was someone on YouTube - that a cheap way

to get fabric for crafts was to pick up the kids' costumes, cuz obviously you just take

the fabric off and use it.

So when I saw these little skirts, I did that.

There's one that's just orange, and then there's one that's black with orange trim.

Again, these were $2.00 each, and again I'm going to use these for embellishments and

things, so I'm excited to get playing with that.

And then also for cheap fabric purposes, I picked up this really gorgeous Halloween scarf.

It was $2.50, and it's just purple with these skull & crossbones on it.

I just thought it was really gorgeous... see if I can show you the... like it's nice, thin

fabric... so again, that should be fun to use for embellishments.

They also had it in red, but I didn't like as much as the purple one.

So I picked that up, again, just for cheap fabric.

I, of course, picked up a bunch of stickers that I did not see when I was at Dollarama

a couple of weeks ago.

I got these googly-eyed ones with the skull wearing a hat, and the owl, and the mummy

- which kills me, because... as you might be able to see, it has one big eye, and one

extremely tiny eye [laughs], and that made me laugh really hard - and there's a vampire

and another skull wearing a bowtie.

So I got two sets of those.

I got these googly-eyed ones with the Frankenstein; again, he's got odd-sized eyes... so does

the zombie... the cat... and a witch... and actually, there's eyeballs in the witches'

brew as well!

Again, thought those were really cute, got two sets of those....

I believe they were each-- these googly-eyed ones were $1.00 each, because there's no sticker on them.

And then I got these sets, which are kinda like the sets you get at Dollar Tree, where

these's like, different things of different sizes... they were $1.50 each.

I did see these at the Dollarama I went to a couple of weeks ago, but they were still

bundled in a big plastic package, like to be put out?

Someone had tossed them into one of the empty baskets.

And I was going to take some out, but then I'm like, "I don't wanna screw up their inventory",

cuz I've worked in retail, you don't want to screw up their inventory and they think

that something's been stolen.

So I was happy to see these ones out where you could buy them!

So I got this one with the ghosts and the owls.

See there, there's like, ghosts... well, ghosts and owls!

They're clear, and... are they puffy?

Yeah, they're a little bit puffy as well.

And then there's glittery ones along the same theme, and then there's... normal ones, I guess?

I don't know what you'd call them, but they're normal... and I didn't realize til I got home

that there's more on the back as well, in little squares, so that's cute.

So I got two packs of those.. again these were $1.50 each.

And then I got these ones as well... again there's clear/puffy ones, or whatever you

want to call them, on the front there... there's skeletons and zombies and all sorts of little things...

And then there's some glitter stickers... and then just normal stickers on the same

theme... and again there's some square ones on the back.

So I got two sets of those as well.

And the last Halloween item I picked up was this wooden sign, it says "Boo" and it has

a skeleton and spider and spiderwebs, it's a little bit glittery.

I just thought it was really cute and didn't want to leave it behind, and it was $2.50.

There were a couple other signs that I liked, but they were all like $3.00-$4.00, so this

one was only $2.50, so I thought I would grab it, and it was the only one there, so... didn't

wanna regret not picking it up.

And then on a slightly more boring note, I just picked up some more resealable craft

bags, cuz again I've been using them to organize stuff... so this was $1.25, there's 72 of

the small...

I thought I would just pick them up at Dollar Tree, but the Dollar Tree bags are actually

smaller than these - the small size that they have there - and this is about the right size

that I need, so... grabbed those.

And then I also grabbed the biggest size, cuz they didn't have-- I could have sworn

they had a medium size, but this store didn't have them.

They were $1.25 as well, 36 in a bag.

And then I finally bought (bit) the bullet and got a glue gun and some glue sticks.

Basically because some of the stuff I want to do, I think it's not going to work too well

with just the glue runners and stuff, so...

I've never owned a glue gun, I'm not really sure if it's gonna work.

I did see-- I popped into the Your Dollar Store With More, but they didn't have anything,

really, that I wanted to pick up, but they did have a glue gun stand.

And I hadn't gone to Dollarama yet, so I didn't pick it up.

So I might have to go back and see if they have the stand, because I don't think this

has a stand on it, which is unfortunate.

But yeah... the glue gun was $3.50 and the sticks were $1.25.

So I thought I would give those a try.

That's all for this haul.

Like I said, it wasn't that big, I only spent, I think about $25-$30 this time around...

which is good!

Considering how much I've been spending at dollar stores and stuff lately.

Sarah Huckabee Sanders has proven she's the woman for the job.

As Press Secretary she's gained even the respect of some of the anti-Trump liberal

media for her honesty and clarity.

Now IJR has a story about her press briefing today on Friday, where Huckabee Sanders smiled

while she was up at the podium and CNN's idiotic Jim Acosta wanted to know why.

Acosta asked Huckabee Sanders about the Madman Theory which is the idea that an effective

leader must keep his enemies guessing and wondering if he is crazy to throw them off.

Trump says "a lot of unsettling off-putting comments that sort of throw people off, that

he likes to keep his adversaries guessing," Acosta said.

Is this an intentional strategy.

Huckabee Sanders looked at him skeptically from the podium, saying of course it is and

POTUS "doesn't want to lay out his game plan for our enemies."

Acosta then followed up, asking how people should react when folks in Washington and

the media "hold their breath" when they hear comments like Trump's yesterday that

this is "the calm before the storm."

"So when people are — they sort of catch their breath in this town, when they sort

of hold their breath in this town, when he says something like that… you have a smile

on your face.

Is that somewhat satisfying?"

Acosta asked.

Huckabee Sanders smiled and absolutely decimated this clown.

Why was she smiling?

"I picture people in this town holding their breath.

That might be a welcome surprise for most of America."

Mic drop.

Boom.

Game over.
